摘 要:建立了拟除虫菊酯类农药多残留酶联免疫检测方法。用不同方法和反应比合成和筛选包被抗原,优化间接竞争ELISA方法反应条件,建立甲氰菊酯标准抑制曲线,测定11种菊酯的交叉反应率。结果显示,以混合酸酐法合成,反应摩尔比45︰1的MA2为最佳包被抗原。优化条件:助溶有机溶剂为甲醇,添加体积分数为10%,离子强度为10mmol/L,pH为7.4。该方法对8种拟除虫菊酯具有较高的灵敏度,IC50范围在8.69~344.97 ng/mL之间。包被抗原的合成条件对于间接竞争ELISA有重要影响。该试验成功建立了可检测多种拟除虫菊酯的间接竞争ELISA方法。To establish an indirect competitive enzyme-linked immunosorbent assay(ELISA)for multiple detection of pyrethroid residues,coating antigens were synthesized via different methods and reaction ratios,and the optimal one for the development of ELISA was screened.The standard inhibition curve of fenpropathrin was established with the optimal reaction conditions.The cross reaction of 11 pyrethroids was determined to verify the multiple detection.The results showed that the optimal coating antigen was MA2,which was synthesized via the mixed anhydride method with the reaction molar ratio of 45︰1.The optimal reaction conditions were as follows:methanol was used for dilution of nonpolar pyrethroids with the volume fraction of 10%.10 mmol/L and pH 7.4 were chosen to be the optimal ionic strength and pH,respectively.The established method could identify 8 pyrethroids with IC50 ranging from 8.69 ng/mL to 344.97 ng/mL.The synthesis conditions of coating antigen had an impact on the results of ELISA.An indirect competitive ELISA method for the simultaneous detection of various pyrethroids was successfully established.
