摘  要:目的研究融合基因重组卡介苗(recombinant BCG,rBCG)对EB病毒阳性肿瘤的作用。方法构建荷瘤小鼠模型,对重组BCG免疫荷瘤小鼠后成瘤时间、肿瘤重量、小鼠存活情况进行分析,利用流式细胞仪测定CD4+T、CD8+T细胞增殖情况,采用乳酸脱氢酶法分析细胞毒性T淋巴细胞(CTL)反应,对免疫后肿瘤组织进行形态学观察,分析淋巴细胞浸润情况。结果当效靶细胞比为50∶1时,PBS组、BCG组和重组BCG组对肿瘤细胞的杀伤率分别为(20.8±1.6)%、(30.8±2.1)%和(60.8±2.3)%。与对照相比,重组BCG对肿瘤免疫效果明显小鼠肿瘤重量明显减小、成瘤时间延迟,CD4+T、CD8+T细胞增殖明显;CTL作用效果增强,肿瘤组织有明显的淋巴细胞浸润。结论重组BCG在小鼠体内产生特异性免疫反应,对肿瘤细胞具有明显的抑制作用。Objective To study the immunological effect of recombinant BCG(rBCG)with a fusion gene on EB virus-positive tumors.Methods A tumor-bearing mouse model was created.Tumor formation time,tumor weight,and the survival of mice immunized with recombinant BCG were analyzed,the proliferation of CD4^+T and CD8^+T cells was determined usingby flow cytometry,the cytotoxic T-lymphocyte(CTL)reaction was analyzed using the lactate dehydrogenase method,the morphology of tumor tissue was observed after immunization,the infiltration of lymphocytes was analyzed,and the immunological effect of recombinant BCG was analyzed statistically.Results When the ratio of effector cells:target cells was 50:1,tumor cells were killed at a rate of 20.8±1.6%in the PBS group,30.8±2.1%in the BCG group,and 60.8±2.3%in the recombinant BCG group were(20.8±1.6)%,(30.8±2.1)%and(60.8±2.3)%,respectively.Recombinant BCG had a significant effect on tumor immunity compared to that in the control group,the tumor weight in mice decreased significantly,tumor formation was delayed,CD4^+T and CD8^+T cells proliferated markedly,the effect of CTL was enhanced,and tumor tissue had marked lymphocyte infiltration.Conclusion Recombinant BCG had a marked immune effect in mice and hada marked inhibitory effect on tumor cells.
