摘  要:目的绿茶中茶多酚和茶黄素成为近年来防龋研究的热点,文中研究茶多酚和茶黄素对3种致龋菌生长作用的影响。方法测定茶多酚和茶黄素对3种致龋菌(变形链球菌、远缘链球菌和粘性放线菌)的最低抑菌浓度(MIC)和最低杀菌浓度(MBC)。测定纯度为20%、40%、60%、80%的茶黄素和纯度为95%的茶多酚培养液对3种致龋菌生长曲线的影响。根据茶黄素和茶多酚对实验菌株的MIC测定结果,选取MIC值以下4个浓度梯度配制BHI液体培养基,实验设阴性对照和阳性对照。阴性对照为不含药物的培养基中的细菌数,阳性对照组为含0.05%洗必泰的培养基中的细菌数。精确称量洗必泰,制备成浓度为0.05%的溶液,并置于4°C冰箱中备用。将茶多酚和茶黄素分别按2倍稀释法溶于BHI液体培养基中,使药物浓度分别为0.25、 0.5、1、2、4、8、16、32、64、128 mg/mL,测定茶多酚和茶黄素对变形链球菌、远缘链球菌和粘性放线菌生长的影响。结果茶多酚和茶黄素对3种细菌的MIC值(mg/mL)为1.0~16.0,MBC值(mg/mL)为4.0~64.0。与空白对照相比较,药物处理后细菌生长对数期的A值差异有统计学意义(P<0.05)。茶多酚和茶黄素对变形链球菌、远缘链球菌和粘性放线菌均有抑制作用,且在细菌的生长对数期的抑制作用最为明显,抑制作用随浓度的增加而增大。茶多酚和茶黄素对变形链球菌和粘性放线菌的抑制作用主要发生在细菌生长前24 h,对远缘链球菌的抑制作用主要发生在细菌生长前16 h,均在细菌对数生长时期。结论茶多酚和茶黄素可抑制口腔致龋菌的生长,能有效调节口腔生态平衡。Objective The present study aims to investigate the effect of tea polyphenols and theaflavins on the growth of three cariogenic bacteria. Methods We determined the MIC and MBC of three kinds of cariogenic bacteria(Streptococcus mutans, Streptococcus sobrinus and Actinomyces viscosus) with tea polyphenols and theaflavins. We also determined the effect of different concentrations of drug-containing culture media on the growth curves of three cariogenic bacteria. Results The MIC range of three kinds of tea polyphenols and theaflavins for three given bacteria was 1.0-16.0 mg/mL, and the MBC value was 4.0-64.0 mg/mL. It showed the best bacteriostatic effect on three cariogenic bacteria when Tea polyphenols with a purity of 95% and theaflavins with a purity of 40%. However, the effect was relatively poor if theaflavins with the purity of 20%, 60%, and 80%. Conclusion Tea polyphenols and theaflavins have strong antibacterial activity against three cariogenic bacteria, and the inhibitory effect increases with the increase of the concentration.
