摘 要:对火麻仁油的多酚提取及福林酚(Folin-Ciocalteu,FC)分光光度法测定总酚含量的条件进行优化,优化后的方法为:吸取0.5 mL 80%乙醇溶液加入到0.25 mL油样中,在室温下(25℃)混匀,以4000 r/min离心15 min,提取3次后合并上相定容至1.5 mL。以没食子酸为标准品绘制标准曲线,火麻仁油多酚提取液中加入0.1 mol/L福林酚试剂3.5 mL,反应10 min后加入7.5%碳酸钠溶液2.5 mL,暗处反应1 h并于765 nm处测定吸光度。对优化方法以没食子酸为标准品进行评价,结果表明没食子酸浓度在10μg/mL~80μg/mL与吸光度的线性关系良好(r2=0.9986),该方法测得加标回收率为(104.41±1.98)%,同一样品重复测定6次的相对标准偏差(relative standard deviation,RSD)为2.20%,0~3 h内测定吸光度的RSD为0.92%,重复性和稳定性良好,可作为火麻仁油的酚类油脂伴随物含量的有效的分析测定方法。The extraction of polyphenols from hemp seed oil and folin-ciocalteu(FC)method for the determination of total phenolic content were optimized.The optimized method was as follows that 0.5 mL 80%ethanol solution was added to 0.25 mL oil sample,which was mixed at room temperature(25℃)and centrifugated at 4000 r/min for 15 min,and then extracted for three times and combined with upper phase volume to 1.5 mL.The standard curve was drawn with gallic acid as the standard,3.5 mL 0.1 mol/L folin phenolic reagent was added to the polyphenolic extract of hemp seed oil,and 2.5 mL sodium carbonate solution with 7.5%mass concentration was added after the reaction for 10 min.The absorbance was measured at 765 nm after the reaction in dark for 1 h.This optimized method was evaluated with gallic acid as a standard,the results showed that gallic acid had a good linear relationship with the absorbance within the mass concentration range of 10μg/mL to 80μg/mL(r2=0.9986).The recovery rate of the method was(104.41±1.98)%,the relative standard deviation(RSD)of the same sample was 2.20%for 6 times determination,and 0.92%for absorbance within 0-3 h,with good repeatability and stability,which could be used as an effective method for the analysis and determination of the content of phenolic oil concomitant in hemp seed oil.
