摘  要:目的对2019年云南昆明地区手足口病患者的粪便进行病毒分离并对毒株进行全基因组序列测定和遗传特性分析。方法利用人类横纹肌肉瘤细胞(human rhabdomyosarcoma,RD)进行病毒分离,提取病毒的RNA序列进行逆转录聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)并测序,利用MEGA 6.06、Simplot 3.5.1和Geneious 9.1.4等软件对毒株的全基因组序列进行分析。结果 KR3/YN/CHN/2019(简称KR3)和KR110/YN/CHN/2019(简称KR110)分离株经测序比对为柯萨奇病毒A组4型(coxsackievirus A,CV-A4),全基因序列长度分别为7 434个核苷酸和7 436个核苷酸,编码2 201个氨基酸;全基因核苷酸一致性及氨基酸同源性分别为92.4%和98.4%。2株CV-A4云南分离株与大多数中国流行株在同一分支,均属于C2亚型。结论 KR3和KR110分离株为CV-A4,与中国CV-A4流行株一致,同属于C2亚型。Objective To analyze the genomic sequence and genetic characteristics of strains isolated from the feces of children with hand, foot and mouth disease(HFMD) in Kunming of China in 2019. Methods RD cells were used to isolate viruses, and viral RNA was extracted.The whole genome sequences of the viruses were amplified by RT-PCR.MEGA6.06, Geneious 9.1.4, and Simplot 3.5.1 software was used to analyze the whole genome sequence of this strain. Results KR3/YN/CHN/2019(KR3) and KR110/YN/CHN/2019(KR110) were classified as type CV-A4.The length of the whole genome sequence of the two strains was 7 434 nt and 7 436 nt, respectively, encoding a polyprotein containing 2 201 amino acid residues.The nucleotide and amino acid homologies between the two strains were 92.4% and 98.4%, respectively.The two Yunnan isolates of CV-A4 and most CV-A4 Chinese isolates belonged to the same genotype, and both grouped into the C2 subtype. Conclusions KR3 and KR110 are CV-A4 strains, which are consistent with the epidemic strains of CV-A4 in China and belong to the C2 subtype.
