摘 要:[目的]建立正相高效液相色谱(NP-HPLC)同时测定万寿菊中玉米黄质和叶黄素含量的方法。[方法]色谱柱Inertsil SIL-100A(4.6 mm×250 mm,3μm),流动相为正己烷∶乙酸乙酯=70∶30(V∶V),检测波长446 nm,柱温30℃。[结果]玉米黄质在5~50μg/mL呈线性关系(r=0.997),加标回收率为97.5%(n=5),检出限为0.055μg/mL;叶黄素在10~100μg/mL呈线性关系(r=0.999),加标回收率为97.3%(n=5),检出限为0.085μg/mL。[结论]建立的正相HPLC方法快速、稳定,能够为检测万寿菊中叶黄素和玉米黄质含量提供有效方法。[Objective]To establish a method for the simultaneous determination of lutein and zeaxanthin in marigold by normal phase high performance liquid chromatography(NP-HPLC).[Method]The column,Inertsil SIL-100A(4.6 mm×250 mm,3μm),mobile phase,n-Hexane∶ethyl acetate=70∶30(V∶V),detection wavelength 446 nm,column oven temperature 30℃.[Result]Zeaxanthin showed a linear relationship between 5-50μg/mL(r=0.997),the recovery rate was 97.5%(n=5),the detection limit was 0.055μg/mL,lutein showed a linear relationship between 10-100μg/mL(r=0.999),the recovery rate was 97.3%(n=5),the detection limit was 0.085μg/mL.[Conclusion]The established HPLC method is rapid and stable,and can provide an effective method for the determination of lutein and zeaxanthin in marigold.
