摘  要:从青藏高原土壤中筛选出一株产木聚糖酶较高的菌株Neurospora crassa SD10。通过单因素试验和正交试验对菌株进行发酵条件优化和酶学性质探究。结果表明,菌株SD10最佳发酵条件为:麸皮30 g/L、蛋白胨20 g/L、Mn^2+0.5 g/L、培养温度33℃、转速230 r/min、接种量2%、初始pH值8.0、发酵时间84 h。菌株产木聚糖酶的最适反应温度为50℃,最适反应pH值为6.0,在40~50℃和pH值4.0~8.0时有较好的稳定性,能保持80%以上酶活力,Fe^2+、Zn^2+、K+对酶活力有促进作用,Na^+、Mg^2+、Ca^2+、Mn2^+、Cu^2+对酶活力有抑制作用。Neurospora crassa SD10优化后产木聚糖酶活力达9.4 U/mL,所产木聚糖酶具有良好的pH值稳定性。Neurospora crassa SD10 was the strain with high xylanase activity from the alpine mead⁃ow soil on the Tibetan Plateau.The fermentation condition and enzymatic properties were conduct⁃ed by single factor and orthogonal test.The re⁃sults showed that the optimum fermentation conditions were as follows:bran 30 g/L,peptone 20 g/L,Mn2+0.5 g/L,33℃,pH 8.0,inoculum amount 2%,rotation speed 230 r/min after 84 h.The enzyme was optimally active at temperature 50℃and pH 6.0.The enzyme activity was relative stable under 40~50℃or pH 4.0~8.0.The xylanase activity of SD10 was enhanced by Fe2+,Zn2+,K+,but reduced by Na+,Mg2+,Ca2+,Mn2+,Cu2+.The highest xylanase activity of Neurospora crassa SD10 reached 9.4 U/mL by optimization.And the xylanase have good pH stability.
