摘  要:为提高生物转化法的木糖醇转化率,促进绿色生产,以生物转化木糖醇的菌株为出发菌株,采用亚硝酸钠和紫外线诱变处理及二轮复合诱变,对高转化木糖醇的菌株进行筛选。在此基础上,通过单因素实验和正交试验,对发酵培养基的氮源和无机盐进行优化。结果获得1株高产菌株,其木糖醇转化率从出发菌株的36.5%提高到54.5%。对该菌株进行发酵培养基的优化,其最佳氮源、无机盐及其质量浓度为:牛肉膏10g/L、蛋白胨20 g/L、硫酸镁0.5 g/L、磷酸二氢钾0.5 g/L。此时菌株木糖醇转化率提高到63.0%。In order to improve the xylitol conversion rate of the biotransformation method and promote green produc-tion,the strains with high xylitol conversion were screened by sodium nitrite and ultraviolet mutagenesis and two-round compound mutagenesis.On this basis,the nitrogen source and inorganic salt of the fermentation medium were optimized by single factor experiment and orthogonal experiments.The xylitol conversion rate of one high-yielding strain was in-creased from 36.5%to 54.5%.The fermentation medium was optimized,and the optimal nitrogen source,inorganic salt and its concentration were:beef extract 10 g/L,peptone 20 g/L,magnesium sulfate 0.5 g/L,potassium dihydrogen phos-phate 0.5 g/L.Under above conditions,the strain's xylitol conversion rate increased to 63.0%.
