摘 要:目的研究大蒜素对脑出血大鼠脑组织的保护作用及其机制。方法将240只SD大鼠随机分为假手术组、脑出血组、大蒜素5 mg/(kg·d)组、大蒜素10 mg/(kg·d)组、大蒜素20 mg/(kg·d)组、尼莫地平组2 mg/(kg·d)组,每组40只。除假手术组外,其余组大鼠采用自体血注射法复制脑出血模型。造模成功后,假手术组和脑出血组给予0.9%氯化钠溶液2 mL/kg腹腔注射,其余组给予相应剂量药物腹腔注射,均1次/d,连续7 d。采用mNSS评分法评估大鼠神经功能缺损程度,伊文思蓝渗透法测定血脑屏障通透性,干湿比重法测定脑组织含水量,通过HE染色法行脑组织病理学观察,TUNEL法检测细胞凋亡情况,黄嘌呤氧化酶法测定脑组织中超氧化物歧化酶(SOD)活性,高锰酸钾滴定法测定脑组织中过氧化氢酶(CAT)活性,硫代巴比妥酸法测定脑组织中丙二醛(MDA)含量,ELISA法测定脑组织中炎症细胞因子肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)含量,Western blot法检测脑组织中核因子-κB(NF-κB)蛋白表达情况。结果脑出血组大鼠mNSS评分、脑组织伊文思蓝含量、脑组织含水量、细胞凋亡指数均显著高于假手术组(P均<0.05);大蒜素10 mg/(kg·d)组、大蒜素20 mg/(kg·d)组和尼莫地平2 mg/(kg·d)组mNSS评分、脑组织伊文思蓝含量、脑组织含水量、细胞凋亡指数均显著低于脑出血组(P均<0.05),且大蒜素20 mg/(kg·d)组mNSS评分、脑组织伊文思蓝含量、细胞凋亡指数均显著低于尼莫地平2 mg/(kg·d)组(P均<0.05)。大蒜素各组和尼莫地平2 mg/(kg·d)组脑组织病理学明显轻于脑出血组,以大蒜素20 mg/(kg·d)组减轻最为显著。脑出血组SOD、CAT活性显著低于假手术组(P均<0.05),MDA、TNF-α、IL-1β、IL-6含量及NF-κB蛋白表达水平均显著高于假手术组(P均<0.05);大蒜素10 mg/(kg·d)组、大蒜素20 mg/(kg·d)组和尼莫地平2 mg/(kg·d)组SOD、CAT活性显著高于脑Objective It is to study the effect and mechanism of Allicin on brain tissue of rats with cerebral hemorrhage.Methods 240 SD rats were randomly divided into sham operation group,cerebral hemorrhage group,allicin 5 mg/(kg·d)group,allicin 10 mg/(kg·d)group,allicin 20 mg/(kg·d)group,nimodipine group 2 mg/(kg·d)group,40 rats in each group.Except for the sham operation group,the rats in the other groups were injected with autologous blood to replicate cerebral hemorrhage models.After successful modeling,the sham operation group and the cerebral hemorrhage group were given an intraperitoneal injection of 0.9%sodium chloride solution 2 mL/kg,and the other groups were given an intraperitoneal injection of the corresponding dose of drugs,all once a day for 7 consecutive days.The mNSS scoring method was used to evaluate the degree of neurological deficits in rats,the Evans blue penetration method was used to measure the permeability of the blood-brain barrier,the dry-wet specific gravity method was used to measure the water content of brain tissue,the pathological change of brain tissue was examined by HE staining and the apoptosis was observed by TUNEL method.The activity of superoxide dismutase(SOD)in the brain tissue was determined by xanthine oxidase method,the activity of catalase(CAT)in the brain tissue was determined by potassium permanganate titration method,and the content of malondialdehyde(MDA)in the brain tissue was detected by thiobarbituric acid method,the contents of the inflammatory cytokines tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6)in brain tissue were detected by ELISA,the expression of NF-κB was determined by Western blot.Results The mNSS score,Evans blue content in brain tissue,water content of brain tissue,and cell apoptosis index in the rats of cerebral hemorrhage group were significantly higher than those in the sham operation group(all P<0.05),the above indexes in the allicin 10 mg/(kg·d)group,20 mg/(kg·d)group and nimodipine 2 mg/(kg·d)group were significa
