摘  要:目的探讨三氧化二砷(As2O3)对人三阴性乳腺癌细胞株MDA-MB-231教化单核细胞的影响。方法于体外分别用不同浓度的As2O3处理MDA-MB-231细胞,采用酶联免疫吸附试验(ELISA)法检测巨噬细胞集落刺激因子1(CSF1)表达水平的差异,利用Transwell共培养体系检测不同浓度As2O3处理组MDAMB-231细胞对人单核细胞株U937趋化能力的差异。结果As2O3能下调MDA-MB-231细胞CSF1的表达,且呈现浓度依赖性(P<0.01);Transwell共培养体系显示,As2O3能减弱MDA-MB-231细胞对人单核细胞株U937的趋化作用。结论As2O3可通过下调CSF1的表达而抑制三阴性乳腺癌细胞株MDA-MB-231对人单核细胞株U937的趋化作用,且呈浓度依赖性。Objective The effects of arsenic trioxide(As2O3)on human triple negative breast cancer cell line MDA-MB-231 educating monocytes was investigated.Methods MDA-MB-231 cells were treated with different concentrations of As2O3 in vitro.The expression of macrophage colony-stimulating factor 1(CSF1)protein was detected by enzyme linked immunosorbent assay(ELISA),and the chemotaxis of MDA-MB-231 cells to human monocyte U937 was detected by Transwell co-culture system.Results As2O3 could down-regulate the expression of CSF1 in MDA-MB-231 cells in a concentration dependent manner(P<0.01).Transwell co-culture system showed that As2O3 could attenuate the chemotaxis of MDA-MB-231 cells to human monocyte U937.Conclusion As2O3 could inhibit the chemotaxis of triple negative breast cancer cell line MDA-MB-231 to human monocyte cell line U937 by down-regulating the expression of CSF1 in a concentrationdependent manner.
