脱除生鲜湿面褐变产物中蛋白质与糖类杂质

摘  要:利用Sevag法和透析法脱除生鲜湿面褐变产物粗提物中的蛋白类杂质,利用AB-8大孔树脂脱除粗提物中的糖类杂质。结果显示:Sevag溶液脱除6次效果较好,蛋白类杂质清除率可达78.08%,经透析后其中蛋白类杂质清除率达到94.00%;使用AB-8大孔树脂脱除糖类杂质,优化后上样参数为母液pH 3、上样流速3 mL/min,洗脱参数为洗脱液为体积分数80%乙醇溶液、洗脱液pH 3、洗脱流速3 mL/min。在此条件下,糖类杂质清除率达到99.74%,褐变产物总回收率为27.23%。经过此方法可有效脱除褐变产物粗提液中蛋白质和糖类杂质,实现纯化褐变产物的目标。Sevag method and dialysis method were used to remove protein impurity from browning products of fresh and wet noodles,and AB-8 macroporous resin was used to remove carbohydrate impurity.The results showed that the best protein removal rate was 78.08%after 6 times removing of Sevag solution.After dialysis,in the browning products,the protein impurity clearance rate was 94.00%.AB-8 macroporous resin was used to remove carbohydrate impurity,and the optimized loading parameters were as follows:mother liquor pH 3,sample flow rate 3 mL/min;elution parameters were as follows:eluent volume fraction 80%ethanol solution,eluent pH 3,elution flow rate 3 mL/min.Under these conditions,the carbohydrate impurity removal rate was 99.74%,and total recovery rate of browning products was 27.23%.The three methods mentioned above could effectively remove protein and carbohydrate impurities from the crude extract of browning products and achieve the goal of purifying browning products.
