摘 要:目的研究乳铁蛋白(lactoferrin,LF)对大鼠上颌扩弓及复发过程中腭中缝破骨活动的影响,并初探其作用机制。方法对大鼠扩弓及复发模型给予剂量为1g/kg/d的LF灌胃,用免疫组织化学染色的方法观察LF对腭中缝破骨活动的影响,检测OPG/RANKL/RANK作用轴关键因子的表达变化。结果与单纯扩弓组相比,LF灌胃组腭中缝区域破骨活动相对减弱,骨保护素(osteoprotegerin,OPG)与核因子KB受体活化因子配体(receptor or activator of nuclear factor-KB ligand,RANKL)表达量的比值显著增加。结论 1g/kg/d LF灌胃可以在大鼠上颌扩弓及复发的过程中抑制腭中缝的破骨活动,此作用可能是通过对OPG/RANKL/RANK作用轴的调控实现的。Objective To evaluate the effects of lactoferrin(LF)on rat bone resorption of the mid-palatal suture during maxillary expansion and relapse,and to preliminarily explore the mechanism.Methods The rat maxillary expansion and relapse model was gavaged daily with LF at the dose of 1 g/kg/d.Immunohistochemical staining was conducted to assess the effects of LF on bone resorption of mid-palatal suture.The expressions of key factors in the osteoprotegerin(OPG)/receptor or activator of nuclear factor-KB ligand(RANKL)/RANK axis were detected.Results Compared to the expansion only group,bone resorption was inhibited in the mid-palatal suture of the LF intervention group,accompanied by significantly increased expression ratio of OPG/RANKL.Conclusion LF gavage at 1 g/kg/d can suppress rat bone resorption of the mid-palatal suture during maxillary expansion and relapse,probably by regulating the OPG/RANKL/RANK axis.
