摘 要:目的:探讨PARP1、HSP70和HSPA8在甲磺酸伊马替尼(imatinib mesylate, IM)治疗的慢性髓系白血病(chronic myelogenous leukemia, CML)患者中的表达水平及其意义。方法:选取2015年1月-2017年12月我院血液科收治的CML患者25例,体检中心健康人群15例,用实时定量RT-PCR(Real Time PCR)方法,对外周血中PAPR1、HSP70以及HSPA8基因表达水平进行测定,并分析PARP1、HSP70和HSPA8与不同病期及IM疗效的关系。结果:初诊CML各病期患者与健康人群相比,PARP1和HSP70基因表达水平显著上调(P<0.05),HSPA8基因表达水平无明显差异;CML患者进展期(加速期和急变期)与慢性期相比,PARP1基因表达水平无明显差异,PARP1、HSP70基因表达水平显著升高(P<0.05)。使用IM治疗后有效组与无效组相比,PARP1基因表达水平显著下降(P<0.05),HSP70和HSPA8基因表达水平无明显差异。结论:HSPA8与CML的发病进展无关,HSP70基因的表达水平与CML的严重程度相关,PARP1可能参与IM治疗CML的过程。以PARP1、HSP70相关机制为研究对象,可能为CML的发生发展提供新的理论依据和新的药物靶点。Objective To investigate the expression levels and significance of PARP1,HSP70 and HSPA8 in imatinib mesy⁃late(IM)treated chronic myelogenous leukemia(CML)patients.Methods 25 patients with CML in hematology depart⁃ment and 15 healthy people in the physical examination center were selected from January 2015 to December 2017 in our hospital.The gene expression levels of PAPR1,HSP70 and HSPA8 were measured by real-time quantitative RT-PCR(Real Time PCR),and the relationship between these genes expressing levels and different CML phases and IM efficacy was ana⁃lyzed.Results Compared with healthy people,the expression levels of PARP1 and HSP70 in patients with newly diagnosed CML were up-regulated significantly(P<0.05),while the expression level of HSPA8 was not significantly different from those in healthy people.Compared with chronic phase(CP),the expression levels of PARP1 and HSP70 in progression phase(accelerated phase and blastic phase,AP and BP)were significantly increased(P<0.05).After IM treatment,the gene expression levels of PARP1 was significantly decreased(P<0.05)in effective group,and the expression levels of HSP70 and HSPA8 had no significant difference between the effective group and invalid group.Conclusion HSPA8 is not related with the progression of CML.The expression level of HSP70 gene is related to the severity of CML.PARP1 may be involved in the process of IM treatment of CML.Taking the related mechanisms of PARP1 and HSP70 as the research ob⁃jects may provide a new theoretical basis and a new drug target for CML occurrence and development.
