摘 要:目的本研究旨在对成都地区2~6岁学龄前儿童肠道双歧杆菌结构特点进行初步探索。方法从本地区两所幼儿园2014年4月至5月期间接收的健康幼儿中选取194例2~6岁的学龄前儿童作为目标,收集其粪便标本并对其肠道中常见8种双歧杆菌的含量进行实时荧光定量PCR(Real-time quantitative PCR)定量分析。结果收集的所有粪便标本中均检出双歧杆菌,其中链双歧杆菌(98.97%)和长双歧杆菌(96.39%)的检出比例最高;随年龄不断增加,婴儿双歧杆菌的检出率呈逐年递减变化趋势(P<0.001),其余7种双歧杆菌在不同年龄学龄前儿童中的检出率未出现统计学差异;90%以上的受试者可检出3~6种双歧杆菌,检出4~5种的占50%以上;按照log10为一个计算单位,双歧杆菌在收集的所有粪便标本检验中的菌量平均值为9.34,链双歧杆菌(8.57)、短双歧杆菌(7.20)和婴儿双歧杆菌(7.12)的菌量在不同年龄组间存在统计学差异(P=0.001,P=0.007,P=0.005);受试者粪便中检出的8种双歧杆菌的平均菌量在性别分布上均未出现统计学差异。结论成都地区2~6岁健康学龄前儿童肠道双歧杆菌数量水平较高,菌种结构组成较为丰富,随年龄增加婴儿双歧杆菌呈现递减趋势,性别因素不影响学龄前儿童的双歧杆菌菌量分布。Objective To characterize the species composition of intestinal Bifidobacteria of the preschool children aged 2-6 years in Chengdu.Methods Totally,194 healthy preschool children aged 2-6 years from two kindergartens in the region from April to May 2014 were randomly selected as the target.And their stool specimens were collected.The contents of 8 common types of Bifidobacteria were quantitatively analyzed by Real-time quantitative PCR.Results Bifidobacterium was detected in all collected fecal samples,with the highest detection rates of Bifidobacterium catenulatum(98.97%)and Bifidobacterium longum(96.39%).With the increase of age,the detection rate of Bifidobacterium infantis showed a decreasing trend year by year(P<0.001).There was no statistically significant difference in the detection rates of the remaining 7 typesof Bifidobacteria in preschool children of different ages.More than 90%of the subjects could be detectedwith 3-6 kinds of Bifidobacterium,anddetection of 4-5 species accounted for 50%above.According to log10 as a unit of calculation,the average bacterial count of Bifidobacterium in all stool specimens was 9.34,and the amount of Bifidobacterium catenulatum(8.57),Bifidobacterium breve(7.20)and Bifidobacterium infantis(7.12)in the intestines of preschool children differed from ages(P=0.001,P=0.007,P=0.005),and no apparent changes were observed for other species.The average amount of 8 kinds of Bifidobacteria detected in the feces of subjects showed no statistical difference in gender distribution.Conclusion The number of intestinal Bifidobacteria in healthy preschool children aged 2-6 years in Chengdu is relatively high,and the bacterial composition is relatively rich.The Bifidobacterium infantis shows a decreasing trend with age,while the gender factor is not influential of thepreschool children Bifidobacteria distribution of bacilli counts.
