摘 要:对41个杂交灵芝(Ganoderma lucidum)菌株进行深层发酵培养,评价其菌丝体干重、胞内多糖含量和得率,及胞内多糖分子量分布特征和刺激巨噬细胞释放NO的活性,结果表明:杂交菌株的菌丝体干重、胞内多糖含量和得率与亲本相比有不同程度的差异,与两个亲本相比,菌丝体干重提高的有29个菌株,胞内多糖含量显著增加的有3个菌株,胞内多糖得率显著提高的有7个菌株;不同菌株胞内多糖均主要含有两个组分,且含量差异较大;不同菌株的胞内多糖在200μg·mL^(-1)时均有明显的刺激巨噬细胞释放NO的活性。综合考虑最终筛选出深层发酵用菌株GZ02,其胞内多糖得率最高,为1.34 g·L^(-1)比亲本G0130和G0154分别提高120%和103%,在50 L发酵罐中对GZ02进行小试,发酵168 h后,菌丝体干重达23.14 g·L^(-1),胞内多糖得率达1.43 g·L^(-1)。Through comparison on dry mycelium weight,intracellular polysaccharide content,and intracellular polysaccharide yield,eight hybrid Ganoderma lucidum strains were selected out of 41 candidates.The selected strains were further studied for molecular weight distribution characteristics of intracellular polysaccharides and the effect of them on NO release in macrophage.The results showed that compared with the parental strains,29 of the 41 candidate hybrid strains showed a greater dry mycelium weight,three hybrid strains had a greater intracellular polysaccharide content and seven hybrid strains had a greater intracellular polysaccharide yield.There are two fractions with large content variations in the intracellular polysaccharides of the eight selected hybrid strains.When used at 200μg·mL^(-1),the intracellular polysaccharides of the selected strains stimulated NO release in macrophage significantly.After a comprehensive evaluation,the hybrid strain GZ02 was selected for submerged fermentation in a 50 L bioreactor.The intracellular polysaccharide yield of GZ02 in shake flask cultures was 1.34 g·L^(-1),which was 120%and 103%greater than that of the parental strains G0130 and G0154,respectively.Upon fermentation in the 50 L bioreactor for 168 h,GZ02 yielded 23.14 g·L^(-1)mycelium(dry weight)and 1.43 g·L^(-1)intracellular polysaccharides.
