摘  要:研究旨在筛选分离能够转化甲酸生产蛋白的高效菌株,并对其生理特性和发酵工艺进行研究。试验以5 g/L甲酸钠为碳源的培养基为筛选培养基,以生物量、甲酸利用率、蛋白含量为评价参数筛选获得高效菌株MA5。经16S rDNA序列分析鉴定,命名为副球菌Paracoccus communis MA5。采用单因素分析法对菌株的生理性能进行分析,发现菌株MA5最佳生长温度37℃、初始pH值7、甲酸钠浓度5 g/L和氮源8 g/L酵母粉。采用3 L发酵系统对菌株MA5的发酵工艺进行分析,发现发酵过程中菌株的甲酸消耗速率最高达0.77 g/(L·h)。在发酵末期菌株生物量干重达4.67 g/L,蛋白含量为37%。因此,副球菌MA5能够高效转化甲酸产单细胞蛋白。The study aimed to screen and isolate efficient strains that can use formic acid to produce single-cell protein,and study its physiological characteristics and fermentation process.In the study,the medium with 5 g/L sodium formate as carbon source was used as screening medium,and the efficient strain MA5 was screened and obtained by the evaluation parameters of biomass,formic acid utilization rate and protein content.The strain was identified and named Paracoccus communis MA5 by 16S rDNA sequence analysis.The physiological property of the strain were analyzed by the single factor method,and the results showed that the optimum growth temperature was 37℃,initial pH value was 5,sodium formate concentration was 5 g/L,and nitrogen source of strain MA5 was 8 g/L yeast extract powder.The fermentation process of strain MA5 was analyzed by the 3 L fermentation system.The results showed that the consumption rate of formate reached up to 0.77 g/(L·h) in the fermentation process,the dry weight of biomass reached 4.67 g/L and the protein content was 37% at the end of fermentation.Therefore,P.communis MA5 can efficiently transform formate to produce single cell protein.
