摘 要:目的:探讨甲状腺乳头状癌(PTC)患者微小肿瘤病毒-322(miR-322)、细胞周期蛋白D1(Cyclin D1)的表达与肉瘤滤过性毒菌致癌同源体B1(BRAF)基因V600E突变的关系。方法:选取2019年1月至2020年4月黑龙江省医院收治PTC患者病理样本120例(PTC组)及结节性甲状腺肿(NG)患者病理样本40例(NG组)、正常甲状腺样本40例(NC组),分别检测样本中BRAF(V600E)突变率、miR-322表达、Cyclin D1表达,再分析比较表达,应用Spearman相关系数分析BRAF(V600E)突变率与miR-322表达相关性;应用Kendall’s tau-b相关系数分析,BRAF(V600E)突变率与Cyclin D1表达相关性。结果:PTC、NG、NC组中miR-322的表达水平分别为2.65±0.45、1.00±0.02、1.00±0.01,PTC组明显高于NG组和NC组,差异有统计学意义(F=533.731,P<0.05);PTC组的Cyclin D1表达阳性率(85.00%)明显高于NG组(42.50%)、NC组(0.00%),差异有统计学意义(χ^(2)=28.432,P<0.05),PTC组BRAF突变率(73.33%)明显高于NG、NC组(0.00%、0.00%),差异有统计学意义(χ^(2)=65.185,P<0.05);miR-322、Cyclin D1、BRAF(V600E)突变与PTC患者性别、年龄、肿瘤直径无关,差异无统计学意义(t/χ^(2)=1.477、0.148、0.253,P>0.05)、(t/χ^(2)=0.787、0.493、1.659,P>0.05)、(t/χ^(2)=0.727、0.215、0.070,P>0.05),与PTC肿瘤结节、包膜侵犯、颈部淋巴结转移、临床分期有关(t/χ^(2)=6.306、19.596、18.828,P<0.01)、(t/χ^(2)=4.773、9.944、12.328,P<0.01)、(t/χ^(2)=8.617、16.752、21.420,P<0.01)、(t/χ^(2)=15.766、5.629、5.539,P<0.05);BRAF(V600E)阳性突变率(73.33%)与miR-322表达(2.77±0.42)、Cyclin D1阳性表达(92.05%)呈正相关(r=0.249、P<0.01,r=0.192、P<0.05),BRAF(V600E)突变阳性患者中的Cyclin D1表达阳性率(92.05%)明显高于阴性患者中的表达阳性率(78.13%),差异有统计学意义(χ^(2)=4.413,P<0.05),BRAF(V600E)突变阳性患者miR-322表达水平明显高于阴性患者,差异有统计学意义(t=2.646,P<0.01)。结论:PTC患者存在miR-322与Cyclin D1的高表达,且与BRAF(V600E)�Objective To explore the relationship between the expression of micro-tumor virus-322(miR-322)and cyclin D1(Cyclin D1)in patients with papillary thyroid carcinoma(PTC)and the mutation of B1(BRAF)gene V600E of the murine sarcoma filtering toxin carcinogenic homologue.Methods From January 2019 to April 2020,120 pathological samples of PTC patients(PTC group),40 pathological samples of nodular goiter(NG)patients(NG group)and 40 normal thyroid samples were selected from January 2019 to April 2020(NC group).The BRAF(V600E)mutation rate,miR-322 expression,and Cyclin D1 expression were detected in the samples,and analyzed and compared.Use Spearman correlation coefficient analysis showed that the mutation rate of BRAF(V600E)was correlated with the expression of miR-322.The correlation coefficient analysis of Kendall′s tau-b showed that the mutation rate of BRAF(V600E)was correlated with the expression of Cyclin D1.Results The expression levels of miR-322 in the PTC,NG,and NC groups were 2.65±0.45,1.00±0.02,1.00±0.01,respectively;those in the PTC group were significantly higher than those in the NG group and the NC group(F=533.731,P<0.05).The positive rate of Cyclin D1 expression in the PTC group was 85.00%,significantly higher than 42.50%in the NG group and 0.00%in the NC group(χ^(2)=28.432,P<0.05).The BRAF mutation rate in the PTC group(73.33%)was significantly higher than that in the NG group and 0.00%in the NC group(χ^(2)=65.185,P<0.05).The miR-322,Cyclin D1 and BRAF(V600E)mutations were not correlated with the gender and age of PTC patients,tumor diameter(t/χ^(2)=1.477,0.148,0.253,P>0.05),(t/χ^(2)=0.787,0.493,1.659,P>0.05),(t/χ^(2)=0.727,0.215,0.070,P>0.05),but with PTC tumor nodules,capsule invasion,cervical lymph node metastasis,and clinical staging(t/χ^(2)=6.306,19.596,18.828,P<0.01),(t/χ^(2)=4.773,9.944,12.328,P<0.01),(t/χ^(2)=8.617,16.752,21.420,P<0.01),(t/χ^(2)=15.766,5.629,5.539,P<0.05).The BRAF(V600E)positive mutation rate(73.33%)was positively correlated with miR-322 expression(2.77±0.42)and C
