摘  要:以米糠为原料,乙酸乙酯为溶剂,热回流法提取植物甾醇。以植物甾醇得率为考核指标,在单因素试验的基础上,利用正交试验对植物甾醇提取工艺进行优化,并利用皂化反应、萃取和重结晶对产物进行分离纯化。结果表明,最佳提取工艺为提取温度70℃、提取时间6 h、料液比1∶14(g/mL)、筛网孔径0.125 mm。在此条件下,植物甾醇得率为4.43 mg/g。经皂化反应、萃取初步提纯,植物甾醇含量为12.21%;在此基础上,甲醇作为溶剂,重结晶提纯,甲醇-产物的液料比为3∶1(mL/g),植物甾醇含量为67.31%。该研究为植物甾醇的综合利用提供理论依据。Using rice bran as raw material and ethyl acetate as solvent,phytosterols were extracted by heat reflux method.Using phytosterol yield as the assessment index,based on single factor experiment,the orthogonal test was used to optimize extraction process of phytosterol,and the products were separated and purified by saponification,extraction and recrystallization.The results showed that the optimal extraction process conditions were extraction temperature 70℃,extraction time 6 h,material-to-liquid ratio 1∶14(g/mL)and mesh size 0.125 mm.Under the conditions,phytosterol yield was 4.43 mg/g.After saponification and extraction,phytosterol content was 12.21%.On this basis,methanol was used as solvent to recrystallize,and phytosterol content was 67.31%when the liquid-to-material ratio of methanol-products was 3∶1(m L/g).The study provided a theoretical basis for the comprehensive utilization of phytosterols.
