摘　　要：Exosome,a new kind of major paracrine secretor,is released into the extracellular environment by the outward budding of various types of cells including neural cells and stem cells.The ultracentrifugation was used to separate exosomes from serum and cell culture supernatant of patients and mice for analysis,and exosomes were injected into ventricles in Applpsl transgenic miee and vitreous of rats with optic nerve clamp injury in vivo.The neural cells or retinal ganglion cells cultured in vitro were used to study the molecular mechanism,and single cells sequencing analysis was performed for the retina cells of rats treated with exosomes.In cell models,we found that several cellular miRNA expression levels are correlated with Aβlevels in vitro.These microRNAs directly regulated Aβgeneration in neural cells,and the exosomal microRNAs in supernatant were also decreased in Aβ-treated cells.For in vivo analysis,we also found that these serum exosomal miRNAs are decreased in APP/PS1 mice compared to wild type mice and were correlated with brain Aβlevels.Furthermore,in 60 Alzheimer's disease(AD)patients,we also found that serum exosomal microRNAs were significantly decreased in AD patients comparing to the peoples in control group.
