Conditioned medium for non-small cell lung cancer promotes proliferation and migration of human bone marrow mesenchymal stem cells

摘　　要：To verify the effects of chemokine 8(CXCL-8)secreted by non-small cell lung cancer cells(A549)on human bone marrow mesenchymal stem cells(hBMSC)movement,and analyzes the Akt pathway promotes the molecular mechanism of CXCL-8 in the hBMSC movement.The supernatants of A549 and human normal lung epithelial cells(BEAS-2B)supernatant were extracted,and then hBMSCs were cultured with these two supernatants.Establish control group,A549 supernatant(A549S)group,BEAS 2B supernatant(BEAS-2BS)group and A549S+Akt inhibitors group.The content of CXCL-8 in supernatant was detected by ELISA;the absorbance value(A value),apoptosis rate and cell migration rate of hBMSCs in each group were detected respectively by MTT assay,Annexin V-PI cell apoptosis assay and Transwell cell migration experiment.The expression of Akt and p-Akt in hBMSCs was detected by W estern blot.
