摘  要:【目的】分析不同高压脉冲电场(high voltage pulsed electric field, HPEF)预处理普洱熟茶对SD大鼠体内活性氧(reactive oxygen species, ROS)的影响。【方法】采用DCFH-DA(2,7-dichlorofuorescin diacetate)结合多功能荧光酶标仪检测大鼠肝、肾、心、脑组织内ROS水平的变化,并利用数据分析软件进行统计分析。【结果】与空白对照相比,HPEF预处理组与未预处理组均能降低大鼠肝、肾、心、脑组织内ROS水平,剂量越高ROS水平越低,且除心脏外,其余组织HPEF预处理组对ROS的抑制作用优于未预处理组;HPEF预处理高剂量组(2.0 g·kg^(-1)·bw)处理8周后,肝脏、肾脏和脑组织中ROS水平可分别降低24.51%、63.20%、68.24%,显著高于空白组(P<0.01);中剂量组(1.0 g·kg^(-1)·bw)可以降低肝脏、肾脏和脑组织中ROS水平20.67%、60.41%、64.17%,显著高于空白组(P<0.01);低剂量组(0.5 g·kg^(-1)·bw)可以降低肾脏和脑组织中ROS水平55.91%、57.67%,明显高于空白组(P<0.01)。除空白组外,大鼠肝、肾、心、脑组织内ROS水平随时间的增加而降低。【结论】普洱熟茶可降低大鼠肝、肾、心、脑组织内的ROS水平,HPEF预处理可提高普洱茶的该抑制作用,可为普洱茶功效研究提供新思路。【Objective】The effects of different high voltage pulse electric field(HPEF)pretreatment Pu’er ripe tea on reactive oxygen species(ROS)in SD mice were analyzed.【Method】The changes of the levels of ROS in the liver,kidney,heart and brain of mice were detected by means of DCFH-DA(2,7-dichlorum diacetate),and the data analysis software was used for statistical analysis.【Result】Compared with the blank group,the levels of ROS in mice liver,kidney,heart and brain tissue could be reduced in Pre-treatments group and Un-treatments group,and the higher the dose was,the lower the level of ROS was,and with the exception of the heart,the inhibitory effect of the other tissues Pre-treatments group on the ROS was better than that of the Un-treatments group.With the higher dose(2.0 g·kg^(-1)·bw)of HPEF pretreatment 8 weeks,the levels of ROS in liver,kidney and brain tissues decreased by 24.51%,63.20%,68.24%,which were significantly higher than those in the blank group(P<0.01);With the medium dose group(1.0 g·kg^(-1)·bw),the levels of ROS in liver,kidney and brain reduced by 20.67%,60.41%,64.17%,which were significantly higher than those in the blank group(P<0.01);with the low dose group(0.5 g·kg^(-1)·bw),the levels of ROS in kidney and brain could reduce by 55.91%,57.67%,which were significantly higher than those in the blank group(P<0.01).Except the blank group,the ROS in liver,kidney,heart and brain decreased with time.【Conclusion】Pu’er ripe tea could reduce the ROS levels in liver,kidney,heart and brain of mice,and HPEF Pre-treatments could improve the inhibitory effect of Pu’er ripe tea,which could provide a new idea for the study of the efficacy of Pu’er ripe tea.
