摘 要:以大豆油脱臭馏出物经甲酯化预处理得到的粗甾醇为原料,采用石油醚洗涤、乙醇精制2步结晶法提取高含量植物甾醇.首先筛选出石油醚作为洗涤溶剂,95%(体积分数,下同)乙醇作为精制溶剂.通过单因素试验,分别研究了石油醚洗涤及乙醇精制过程中料液比、养晶温度、养晶时间对甾醇含量及收率的影响,并进行正交试验,对甾醇提取工艺进行优化.结果表明:石油醚洗涤过程最佳条件为料液比1:7(g/mL)、养晶温度26℃、养晶时间2 h,在此条件下甾醇含量为93.5%,收率为96.4%;95%乙醇精制过程最佳条件为料液比1:10(g/mL)、养晶温度30℃、养晶时间4 h,在此条件下甾醇含量达到98.7%,收率89.7%.原料先经过石油醚洗涤,再用95%乙醇精制,最终得到98.7%的高含量植物甾醇.The crude sterols obtained from the soybean oil deodorized distillate through methyl esterification pretreatment were used as raw materials,and the high-content phytosterols were extracted by a twostep crystallization method of petroleum ether washing and ethanol refining.Firstly,select petroleum ether as the washing solvent,and 95%(volume fraction,the same below)ethanol as the refining solvent.Through single-factor experiments,the effects of material-to-liquid ratio,crystal growth temperature,and crystal growth time on the content and yield of sterols in the washing and ethanol refining processes of petroleum ether were studied respectively,and orthogonal experiments were performed to optimize the extraction process of sterols.The results showed that the best conditions for the washing process of petroleum ether were the material-to-liquid ratio 1∶7(g/mL),crystallization temperature 26℃,and crystallization time 2 h.Under these conditions,the sterol content was 93.5%,and the yield was 96.4%;The best conditions for the 95%ethanol refining process were the material-to-liquid ratio 1∶10(g/mL),the crystal growth temperature of 30℃,and the crystal growth time of 4 h.Under these conditions,the sterol content reached 98.7%,and the yield was 89.7%.The raw materials were washed with petroleum ether and then refined with 95%ethanol to obtained 98.7%high-content phytosterols.
