摘 要:神经示踪技术是探索脑内神经元及其连接模式的经典方法。首都医科大学的神经生物学神经示踪实验教学采用辣根过氧化物酶(HRP)逆行示踪舌下神经核运动神经元,以掌握动物麻醉、灌注固定和取材等基本手术操作以及脑组织冰冻切片和组织化学染色等。同时,实验教学讲解了这些环节相关的背景知识,如不同的示踪剂选择和HRP不同底物的组织化学显色方法以及神经示踪技术简史及其在科研中的实际应用等,帮助学生验证并理解轴突转运和神经投射等基础知识,并培养他们将来从事科研工作的兴趣。Neural tracing is a classic method to study neurons and their connection patterns in the brain. In the experiment teaching, horseradish peroxidase(HRP) was used to trace retrogradely the motor neurons of hypoglossal nucleus, so that the undergraduates can master the basic operation skills of animal anesthesia, perfusion, and brain removal, as well as acquire knowledge of brain tissue sectioning and histochemical staining. Besides, the instructors taught operation-related knowledge of the selection of different tracers, histochemical staining of different HRP substrates, the brief history of neural tracing, and its application in scientific research, helping students verify and understand the basic knowledge of axonal transport and neural projection in the brainstem, and cultivate an interest in scientific research.
