摘 要:目的探讨结肠癌转移相关基因-1(metastasis-associated in colon cancer-1,MACC1)在结直肠癌(colorectal carcinoma,CRC)的表达及临床病理意义。方法免疫组织化学检测MACC1在77例CRC及其正常黏膜组织的表达及分析其临床病理意义。HPA和GEPIA数据库分析MACC1在CRC的mRNA和蛋白表达以及与预后的关系,GEPIA数据库分析MACC1表达与错配修复基因的相关性及与CRC临床分期的关系。结果MACC1在CRC的阳性表达率为75.32%,显著高于癌旁正常黏膜的11.69%。MACC1的表达与CRC的淋巴结转移、神经侵犯和临床分期有关,而与患者性别、年龄、肿瘤大小、分化程度、侵犯深度和部位无关。公共数据库分析证实MACC1在CRC表达增加。GEPIA数据库分析显示在不同的临床分期各组间MACC1表达差异具有统计学意义,MACC1基因的表达与错配修复基因MSH2、MSH6、MLH1和PMS2的表达均呈显著相关性。两个数据库分析均显示高表达MACC1的CRC患者生存预后差。结论MACC1 mRNA和蛋白在结直肠癌均高表达,并与结直肠癌侵袭、转移、预后和错配修复基因密切相关,MACC1在结直肠癌恶性进展中起到致瘤作用,具有重要临床价值。Objective To investigate the metastasis-associated in colon cancer-1 (MACC1) expression in colorectal carcinoma(CRC) and its clinicopathological significance.Methods The expression of MACC1 in 77 CRC tissue samples and 77 adjacent normal tissue samples were detected by immunohistochemistry.The role of MACC1 in CRC was analyzed.Public databases including the HPA database and GEPIA database were employed to investigate the expression and prognostic value of MACC1 in CRC.Correlations between the MACC1 mRNA expression and the mismatch repair gene expression and clinical stages of CRC were analyzed with the data from the GEPIA database.Results The positive expression rate of MACC1 in CRC was 75.32%(58/77),while the MACC1 positive expression rate in adjacent normal tissues was 11.69%(9/77).The expression of MACC1 was positively correlated with the lymph node metastasis,nerve invasion,and clinical stage,while had no correlation with the age,gender,tumor size,differentiation degree,invasion depth,and pathogenic site.MACC1 gene expression data from the public databases all proved that MACC1 was overexpressed in CRC.The data from the GEPIA database indicated that the expression of MACC1 was positively correlated with the clinical stages of CRC,and the expression of the MACC1 gene was significantly correlated with the expression of mismatch repair genes MSH2,MSH6,MLH1,and PMS2.According to the analysis of public data,a higher expression of MACC1 was associated with a worse prognosis for CRC patients.Conclusion both MACC1 mRNA and protein are highly expressed in CRC tissues and closely related to the invasion,metastasis,and prognosis of CRC and the expression of mismatch repair genes.MACC1 poses oncogenic effects during the malignant progression of CRC,therefore it has important clinical value.
