摘 要:为研究CsCAT3基因在冷驯化诱导采后黄瓜耐冷性中的作用,采用同源克隆法克隆了CsCAT3基因,并对其基本特性进行分析,研究了冷驯化处理对采后黄瓜CsCAT3表达、CAT活性和过氧化氢(H_(2)O_(2))含量的影响。结果表明,CsCAT3基因的开放阅读框长度为1 476 bp,编码492个氨基酸残基。CsCAT3的理论分子量为57.01 kDa,等电点为6.84,为亲水性蛋白质,无跨膜结构域,定位在过氧化物酶体。CsCAT3与4种模式植物CAT3的同源性介于69.29%-71.82%之间,物种间CsCAT3的同源性和保守性很高。冷驯化处理可明显上调CsCAT3在转录和蛋白水平上的表达,提高CAT活性,降低H_(2)O_(2)含量,表明CsCAT3基因在冷驯化诱导的耐冷性中具有重要作用。本研究为进一步解析CsCAT3在采后黄瓜诱导耐冷性中的功能奠定了基础。To investigate the roles of catalase 3(CsCAT3) gene in pre-storage cold acclimation(PsCA)-induced cold tolerance in harvested cucumber, CsCAT3 was cloned using homologous cloning method, and the basic characteristics was analyzed. Effects of PsCA on CsCAT3 expression, CAT activity and hydrogen peroxide(H_(2)O_(2)) contents were investigated in harvested cucumber. The results showed that the length of open reading frame of CsCAT3 was 1476 bp, which encoded 492 amino acid residues. The theoretical molecular weight and isoelectric point of CsCAT3 were 57.01 kDa and 6.84, respectively. It was a hydrophilic protein with no transmembrane domain and distributed in peroxisomes. The nucleotide homology between CsCAT3 and other CAT3 from four model plants were ranged from 69.29 to 71.82 %. The protein homology and conservation of CAT3 among different plant species were very high. PsCA treatment obviously up-regulated the expression of CsCAT3 at gene and protein levels, enhanced the activities of CAT, but reduced the H_(2)O_(2) levels, suggesting that CsCAT3 played important roles in PsCA-induced chilling tolerance in harvested cucumbers. This study provided basis for understanding the functional roles of CsCAT3 gene in the induction of chilling tolerance in harvest cucumbers.
