摘 要:为探索易逝期贝类活品品质变化机制,以壳长(11.01±0.31)cm的虾夷扇贝Patinopecten yessoensis为研究对象,基于超高效液相色谱-质谱(UPLC-MS)的代谢组学方法研究了其湿藏条件下的代谢特征,试验设活品虾夷扇贝对照组(A)和湿藏组(C),并对虾夷扇贝闭壳肌的pH、糖原及代谢组学进行分析。结果表明:捕后湿藏的虾夷扇贝闭壳肌pH和糖原水平变化不大;经多元统计分析,品质易逝期湿藏36 h的虾夷扇贝闭壳肌中共检测出7个显著性差异代谢物,分别为二十二烷酸(behenic acid)、酪胺(tyramine)、脯氨酰谷氨酸(prolyl-glutamate)、二磷酸腺苷葡萄糖(ADP-glucose)、尿苷5′-二磷酸(UDP)、3-磷酸丝氨酸(3-phosphoserine)和柠檬酸(citrate);通路分析显示,三羧酸循环(TCA cycle)是最易受影响的代谢通路。研究表明,品质易逝期湿藏36 h条件下的活品虾夷扇贝与初始扇贝相比,传统理化指标变化不大,但其代谢组存在显著性差异,说明代谢组学方法可较为灵敏、全面地反映活品虾夷扇贝湿藏期间的实时状态。In order to explore the mechanism of changes in quality of scallop during quality determination period(QDP),pH,glycogen and metabolomics of the adductor muscles were analyzed in post-harvest live Yesso scallop Patinopecten yessoensis with shell length of(11.01±0.31)cm held at 6.9℃(control group),and storage in the water(group C)for 36 h(QDP)at 0,3,6,12,24 and 36 h by an ultra-high performance liquid chromatography-mass spectrometry(UPLC-MS)to analyze the metabolic characteristics of post-harvest Yesso scallop in seawater storage during QDP.The results showed that pH and glycogen contents were found to be slight fluctuation in the Yesso scallop in group C.By multivariate statistical analysis revealed that 7 discrimination metabolites were detected in the Yesso scallop in group C for 36 h,including behenic acid,tyramine,prolyl-glutamate,ADP-glucose,UDP,3-phosphoserine and citrate.The pathway analysis showed that the most easily affected metabolic pathway was TCA cycle.The findings indicated that live Yesso scallops wet stored for 36 h did not change much in physiological characteristics of initial status during quality determination period,compared with the traditional physical and chemical indicators,with significant differences in metabolome,indicating that the metabolomics could be more sensitive and comprehensively reflecting the real-time state of live scallops during wet storage.
