摘  要:胆汁酸(bile acid)是肝脏胆固醇代谢的主要终产物,最近已被证明是控制脂质代谢、能量消耗和葡萄糖稳态的重要信号分子。胆汁酸稳态和胆汁酸介导的信号通路的改变可能影响肝脏代谢稳态,并导致肝胆疾病的发生。质谱(mass spec⁃trometry,MS)检测技术的进步,结合多变量统计方法,为肝胆疾病的早期诊断、靶向治疗和预后提供了一种很有前景的方法。本文首先简要介绍基于质谱的胆汁酸代谢组学技术和数据集分析方法,然后重点综述在常见的几种肝胆疾病中,基于该技术发现的可能用于早期诊断和治疗的特异性代谢标志物,旨在为实现肝胆疾病精准诊疗提供新的理论依据。Bile acids,the main end-products of cholesterol metabolism in the liver,have recently been dem⁃onstrated as important signaling molecules that regulate lipid metabolism,energy expenditure and glucose homeosta⁃sis.Alterations in bile acid homeostasis and bile acid-mediated signaling pathways may affect hepatic metabolic ho⁃meostasis and cause the onset of hepatobiliary diseases.Advanced mass spectrometry,in combination with multivar⁃iate statistical methods,provides a promising approach to early diagnosis,targeted therapy,and prognosis evalua⁃tion of hepatobiliary diseases.This article briefly introduces mass spectrometry-based bile acid metabolomics and dataset analysis methods,and focuses on specific metabolic markers found by this technique that may be used for early diagnosis and treatment in several common hepatobiliary diseases,aiming to provide a new theoretical basis for precise care of hepatobiliary diseases.
