摘 要:旨在研究鸽乳中产纤维素酶及脂肪酶微生物的种类。通过筛选培养基分离出产酶菌株,观察其菌落形态,测定羧甲基纤维素酶、滤纸酶和脂肪酶活力,提取基因组DNA,扩增16S rDNA序列,测序结果经BLAST比对和相似性分析,构建系统发育树。结果:分离出产纤维素酶菌种4个,分别为铜绿假单胞菌(Pseudomonas aeruginosa)、大肠杆菌(Escherichia coli)、吉氏库特氏菌(Kurthia gibsonii)和肺炎克雷伯氏菌(Klebsiella pneumoniae);产脂肪酶菌种1个,为变栖克雷伯氏菌(Klebsiella variicola)。除吉氏库特氏菌外其他均为条件致病性菌。本试验结果将为鸽子消化道微生物多样性和产酶微生物的研究及应用奠定基础。The aim of this experiment was to study the species of cellulose-and lipase-producing microorganisms in pigeon milk. Enzyme-producing bacteria were isolated using the selective culture medium. The bacteria were observed for their colony morphology, and the activities of carboxymethyl cellulase, filter paper enzyme and lipase were determined. Then, genomic DNA was extracted and the 16 S rDNA sequence was amplified. The results of sequencing were compared by blast and similarity analyses, and a phylogenetic tree was constructed. It was shown that four cellulose-producing strains, including Pseudomonas aeruginosa, Escherichia coli, kurthia gibsonii and Klebsiella pneumoniae, and one lipase-producing strain, Klebsiella variicola was identified. All of strains were conditional pathogenic bacteria except Kurthia gibsonii. The results of this experiment laid a foundation further research on and application of microbial diversity and enzyme producing microorganisms in the digestive tract of the pigeon.
