摘 要:目的探讨不同莓果(蓝莓、枸杞及桑葚)提取物对小鼠慢性酒精性肝病(ALD)的作用。方法昆明种小鼠110只随机分为空白组(不做任何干预)、模型组(实验结束前15 d灌胃给予30%乙醇)、蓝莓提取物[25(低)、50(中)及100 mg/kg(高剂量)]组、枸杞提取物[25(低)、50(中)及100 mg/kg(高剂量)]组、桑葚提取物[25(低)、50(中)及100 mg/kg(高剂量)]组,3种莓果提取物低、中、高剂量组小鼠每日灌胃予相应剂量莓果提取物进行预保护,并于实验结束前15 d再灌胃30%乙醇复制慢性ALD模型,实验周期1个月,实验结束时称量各组小鼠体质量;各组小鼠腹腔注射60 mg/kg戊巴比妥钠溶液麻醉,摘眼球取血,检测血清胆固醇(CHOL)、总胆红素(TBIL)、低密度脂蛋白(LDL)、谷丙转氨酶(ALT)及天冬氨酸转氨酶(AST)含量;取各组小鼠肝脏组织计算肝脏指数,同时取各组小鼠肝左叶组织制作切片、苏木精-伊红染色法(HE)观察肝脏组织形态学改变。结果与空白组比较,模型组体质量降低、肝脏指数升高,血清ALT、AST、LDL、TBIL及CHOL含量升高(P<0.05或<0.001);与模型组相比,桑葚提取物低、中、高剂量组小鼠体质量升高、肝脏指数下降(P<0.05);相较于模型组,桑葚、枸杞提取物低、中、高剂量组小鼠血清ALT含量降低(P<0.05);桑葚、枸杞、蓝莓提取物低、中、高剂量组小鼠血清AST、LDL、TBIL及CHOL含量低于模型组(P<0.05);HE染色结果显示,与模型组相比,桑葚提取物高剂量组小鼠肝组织空泡变性明显降低及得以细胞形态大小保护,枸杞及蓝莓提取物低、中、高剂量组小鼠肝细胞水肿、空泡变性、嗜酸变性及中央静脉周围肝细胞水样变性部分降低。结论蓝莓、枸杞、桑葚提取物对小鼠慢性ALD均有保护作用,其中桑葚提取物的作用最佳,其次为枸杞提取物和蓝莓提取物。Objective To investigate the effects of different berry fruit extracts on chronic alcoholic liver disease(ALD)in mice,including blueberry,wolfberry,and mulberry extracts.Methods One hundred and ten Kunming mice were randomly divided into the control group without any intervention.The model group was given 30%ethanol in 15 days before the end of the experiment.The blueberry extract group was given[25(low dose),50(medium dose)and 100 mg/kg(high dose)],the wolfberry extract group with the same dose,and the mulberry extract group with the same dose,respectively.In three groups,different doses of berry extracts were given every day for pre protection,and 15 days before the end of the experiment,30%ethanol was given to cause chronic ALD.The experimental lasted for 1 month.At the end of it,the body weight of mice in each group was weighed;and all mice were anesthetized by intraperitoneal injection of 60 mg/kg pentobarbital sodium solution.The eyeballs were removed to get serum,then the contents of serum cholesterol(CHOL),serum total bilirubin(TBIL),low density lipoprotein(LDL),alanine aminotransferase(ALT),and aspartate aminotransferase(AST)were detected by automatic biochemical analyzer.The liver index was calculated by weighing liver tissue of each group.At the same time,the left lobe tissues of each group were taken to make sections for the pathomorphological observation of liver in each group by hematoxylin and eosin staining.Results Compared with the control group,the body weight of the model group decreased,the liver index increased,and the contents of serum ALT,AST,LDL,TBIL and CHOL increased(P<0.05 or<0.001).Compared with the model group,the body weight of mice in low,medium and high dose groups of mulberry extracts increased and the liver index decreased significantly(P<0.05);Compared with the model group,the content of serum ALT in the low,medium,and high dose groups of mulberry and wolfberry extracts decreased significantly(P<0.05).The contents of serum AST,LDL,TBIL,and CHOL in the low,medium and high dose gr
