摘  要:以酸枣仁脱脂粉为原料,基于Osborne法对酸枣仁组分蛋白进行分离提取,通过单因素试验对酸枣仁蛋白质组分的提取工艺进行优化,并通过SDS-PAGE电泳对酸枣仁组分蛋白进行亚基分析研究。结果表明,酸枣仁清蛋白提取的最佳温度为45℃,球蛋白提取的最佳盐浓度为5%,醇溶蛋白提取的最佳醇浓度为70%,谷蛋白提取的最佳碱浓度为0.1 mol/L。采用最佳提取工艺获得的蛋白提取率为清蛋白13.24%、球蛋白22.25%、醇溶蛋白1.35%、谷蛋白13.57%。SDS-PAGE凝胶电泳结果表明,酸枣仁清蛋白在10~55 kDa均有分布,酸枣仁球蛋白主要分布于10~25kDa,酸枣仁谷蛋白主要分布于15~25 kDa。Based on Osborne method,the protein components of Semen Ziziphi Spinosae were isolated and extracted.The extraction process of protein components of Semen Ziziphi Spinosae was optimized by single factor experiment.The protein components of Semen Ziziphi Spinosae were studied by SDS-PAGE electrophoresis.The results showed that the optimum temperature of albumin was 45℃,the optimum salt concentration of globulin was 5%,the optimum alcohol concentration of gliadin was 70%,and the optimum alkali concentration of glutelin was 0.1 mol/L.The extraction yield of protein was albumin 13.24%,globulin 22.25%,gliadin 1.35%and glutenin 13.57%.The SDS-PAGE gel electrophoresis results showed that jujube albumin was distributed in the range of 10-55 kDa;Semen Ziziphi Spinosae globulin was mainly distributed in the range of 10-25 kDa;Glutelin was mainly distributed in the range of 15-25 kDa.
