摘  要:为探究夏黑葡萄叶片衰老相关基因的表达情况及代谢通路,本研究以叶果比12:1修剪的夏黑葡萄为材料,采用高通量测序技术对采集花后45、65、85和105 d的叶片进行转录组分析。结果显示,花后45 d的样品中得到50855290条clean reads,GC含量为46.49%;花后65 d的样品中得到50432588条clean reads,GC含量为46.13%;花后85 d的样品中得到52169372条clean reads,GC含量为47.10%;花后105 d的样品中得到50988842条clean reads,GC含量为47.01%。与参考基因组进行序列比对,发现各样品74.20%以上的reads比对到参考基因组,绝大部分分布在外显子区,其中有72.43%的reads比对到参考基因组的唯一位置。各样品间基因差异表达分析得到共有差异基因2583个。GO功能注释分析发现这些差异基因主要参与生物过程中的光合作用,以及与核酸结合转录因子活性等分子功能相关;SNP位点分析结果发现在每个样品中平均有417490个变异位点,主要分布在外显子区、内含子区、基因上下游区域。本研究明确了夏黑葡萄叶片衰老过程中相关基因的变化,为进一步挖掘夏黑葡萄叶片衰老过程中关键基因的功能奠定了理论基础。In order to investigate senescence-related metabolic pathways and the expression of genes,the leaves of Summer Black grape were pruned at leaf to fruit ratio of 12:1 in the growing season,and leaves located opposite to fruit were collected at 45,65,85 and 105 days after flowering(DAF)and used for transcriptome analysis by RNA-seq technology.The results showed that.50855290 clean reads were obtained from samples at 45 DAF,with GC content of 46.49%;50432588 clean reads with GC content of 46.13%were obtained from samples at 65 DAF;52169372 clean reads with GC content of 47.10%were obtained from samples at 85 DAF;50988842 clean reads with GC content of 47.01%were obtained from samples at 105 DAF.Results of sequence alignment with the reference genome revealed that more than 74.20%of reads in each sample were aligned to the reference genome,more than 72.43%of which were aligned to unique locations in the reference genome.Most of the sequences aligned to the genome were distributed in the exon region.The analysis of gene differential expressions among samples yielded 2583 common differential genes.GO functional annotation analysis revealed that these differential genes were mainly involved in photosynthesis in biological processes and related to molecular functions such as nucleic acid binding transcription factor activity.Results of SNP mutation sites revealed that an average of 417490 SNP sites were found in each sample,mainly distributed in the exon region,intron region,and upstream and downstream regions of genes.Transcriptome analysis revealed the changes of related genes in the senescence process of grape leaves,which laid a theoretical foundation for further exploring the functions of key genes in the aging process of grape leaves.
