摘　　要：目的探讨DOC-2/DAB2结合蛋白(DAB2IP)、caspase 8在皮肤基底细胞癌(BCC)中的表达及作用机制。方法选取80例BCC患者和30例接受整形外科手术的健康者,分别取其BCC组织和正常皮肤组织,逆转录聚合酶链反应(RT-PCR)检测DAB2IP mRNA的相对表达量,蛋白质印迹(Western blot)法检测DAB2IP、caspase 8蛋白的相对表达量。将si-DAB2IP、si-NC通过脂质体转染至BCC细胞系TE-354.T,分别作为si-DAB2IP组和si-NC组,Western blot法检测细胞中DAB2IP、caspase 8蛋白的相对表达量。结果BCC组织中DAB2IP mRNA的相对表达量明显低于正常皮肤组织(P﹤0.01),DAB2IP、caspase 8蛋白的相对表达量均明显低于正常皮肤组织(P﹤0.01)。抑制DAB2IP基因的表达后,si-DAB2IP组细胞中DAB2IP、caspase 8蛋白的相对表达量均明显低于si-NC组(P﹤0.01)。肿瘤直径≥1 cm、病理类型为浅表型BCC患者BCC组织中DAB2IP、caspase 8蛋白的相对表达量均高于肿瘤直径﹤1 cm、病理类型为浸润型患者(P﹤0.05)。Pearson相关分析结果表明,BCC组织中caspase 8的表达与DAB2IP的表达呈正相关(r=0.72,P﹤0.05)。结论caspase 8、DAB2IP在BCC组织中低表达,DAB2IP可能通过促进caspase 8的表达发挥抗肿瘤作用,靶向DAB2IP或可成为BCC治疗的新靶点。Objective To explore the expression and mechanism of DOC-2/DAB2 interacting protein(DAB2IP)and caspase 8 in skin basal cell carcinoma(BCC).Method A total of 80 cases suffering BCC and 30 healthy subjects undergoing by plastic surgery were selected,and their BCC tissues and normal skin tissues were taken,respectively.Reverse transcription-polymerase chain reaction(RT-PCR)was used to detect the relative expression of DAB2IP mRNA,and Western blot was used to detect the relative expression of DAB2IP and caspase 8 proteins.The si-DAB2IP and si-NC were transfected into the BCC cell line TE-354.T by liposome,and then definite as the si-DAB2IP group and si-NC group,respectively,and the relative expression of DAB2IP and caspase 8 proteins in the cells was detected by Western blot.Result The relative expression of DAB2IP mRNA in BCC tissue was significantly lower than that in normal skin tissue(P<0.01),and the relative expression of DAB2IP and caspase 8 protein was significantly lower than that in normal skin tissue(P<0.01).After inhibiting the DAB2IP gene,the relative expression of DAB2IP and caspase 8 protein in the si-DAB2IP group were significantly lower than that in the si-NC group(P<0.01).The relative expression of DAB2IP and caspase 8 protein in BCC tissues of patients with tumor diameter≥1 cm and pathological type of superficial BCC were higher than those of tumor diameter<1 cm and pathological infiltrating type(P<0.05).The results of Pearson correlation analysis showed that the expression of caspase 8 in BCC tissue was positively correlated with the expression of DAB2IP(r=0.72,P<0.05).Conclusion caspase 8 and DAB2IP are down-regulation in BCC tissues.Notedly,DAB2IP may play an anti-tumor effect by promoting the expression of caspase 8,indicating that DAB2IP may become a promising new target for BCC treatment.
