摘 要:以乙醇为溶剂,采用内部沸腾法提取脐橙皮色素,并利用单因素实验和单因素实验基础上建立的响应面模型对液料比、温度和时间等影响因素进行优化;还初步研究了pH对脐橙皮色素稳定性的影响及测定了脐橙皮色素对羟基自由基的清除率.结果表明,修正后的脐橙皮色素的最佳提取条件为:液料比45∶1 mL/g,温度60℃和时间10 min,此条件下的吸光度的理论预测值为0.335.在此最佳条件下作了5次平行实验,得到吸光度平均值0.345(RSD=4.08%),与理论预测值0.335的相对偏差为2.99%.且此平均吸光度值高于相同条件下有机溶剂直接浸提法时的吸光度0.275,体现了内部沸腾对色素提取的正面促进作用.稳定性实验表明,酸性条件使脐橙皮色素吸光度降低,碱性条件具有一定的增色作用.通过对脐橙皮色素对羟基自由基清除率的测定表明,脐橙皮色素具有一定的抗氧化活性,其清除率可达33.45%.With ethanol as the solvent,pigment in navel orange peel was extracted by inner ebullition method.The influencing factors,such as liquid-solid ratio,temperature,and time,were optimized by aid of single-factor experiments and the response surface model,which was also built on the basis of such single-factor experiments.Influence of pH on stability of pigment in navel orange peel,was studied,and the clearing rate for hydroxyl radical was determined,too.The results showed that the optimal extraction conditions after modification,were liquid-solid ratio at 45:1 mL/g,temperature at 60℃and time duration of 10 min,under which conditions the theoretical value of the predicted absorbency was 0.335.Under the optimal conditions,five parallel experiments were conducted,and the average absorbency obtained was 0.345(RSD=4.08%),which deviated from the relative theoretical value of 0.335 by 2.99%.The average absorbency was higher than the absorbency 0.275 gotten by direct extraction method with organic solvent under the same conditions,which reflected the positive promotion effect of inner ebullition on extraction of pigment.And stability experiments find that the absorbency of pigment weakened in acid conditions,and the color of pigment was enhanced in alkalinity.By determining the clearing rate of the pigment for hydroxyl radical,it indicated that pigment in navel orange peel had some antioxidant activity,and the clearing rate could reach 33.45%.
