摘 要:目的探讨长链非编码RNA HLA-DQB1-AS1调控miRNA-205-5p表达对宫颈癌增殖、迁移和侵袭中的影响。方法检测HLA-DQB1-AS1在宫颈癌及其对应宫颈癌旁组织、H8(人宫颈永生化上皮细胞系)及Siha和Caski(人子宫颈癌细胞系)中的表达;预测并验证HLA-DQB1-AS1、miRNA-205-5p和HLA-DQB1的调控关系;检测Siha和Caski细胞的迁移、侵袭及增殖能力;分析调控HLA-DQB1-AS1表达对人宫颈癌裸鼠皮下移植瘤的影响。结果 HLA-DQB1-AS1在宫颈癌细胞和组织中表达量上调;HLA-DQB1-AS1靶向结合miRNA-205-5p,且HLA-DQB1是miRNA-205-5p的靶基因, HLA-DQB1-AS1通过miRNA-205-5p促进HLA-DQB1的表达;体内外实验证实HLA-DQB1-AS1过表达促进宫颈癌细胞的增殖、侵袭和转移能力(P <0.05),反之起抑制作用;(均P <0.05)。结论 HLA-DQB1-AS1在宫颈癌组织细胞中高表达。HLA-DQB1-AS1通过负调控miRNA-205-5p促进HLA-DQB1表达,促进宫颈癌细胞的增殖、侵袭及迁移。Objective To investigate the effect of long non-coding RNA HLA-DQB1-AS1 regulating the expression of miRNA-205-5 p on the proliferation, migration and invasion of cervical cancer. Methods The level of HLADQB1-AS1 in cervical cancer and its corresponding cervical tissues were detected, H8(human cervical immortalized epithelial cell line) and Siha and Caski(human cervical cancer cell line) were detected;HLA-DQB1-AS1, miRNA-205-5 p and HLA-DQB1 were predicted and verified;the migration, invasion and proliferation of Siha and Caski cells were detected;the effect of regulating expression of HLA-DQB1-AS1 on subcutaneous xenografts of human cervical cancer in nude mice were detected. Results HLA-DQB1-AS1 was up-regulated in cervical cancer cells and tissues;HLA-DQB1-AS1 targeted mi RNA-205-5 p, and HLA-DQB1 was target gene of miRNA-205-5 p. HLA-DQB1-AS1 promoted expression of HLA-DQB1 by miRNA-205-5 p. In vivo and in vitro experiments have confirmed that the overexpression of HLA-DQB1-AS1 promot the proliferation, invasion and metastasis of cervical cancer cells(P < 0.05), and it had an inhibitory effect(all P < 0.05). Conclusion HLA-DQB1-AS1 is highly expressed in cervical cancer tissue cells, HLA-DQB1-AS1 can promot expression of HLA-DQB1 through negative regulation miRNA-205-5 p, and promot the proliferation, invasion and migration of cervical cancer cells.
