摘 要:为了确定盐析法脱除玛卡多糖中的蛋白质的最佳工艺,以多糖回收率与蛋白质清除率为考察指标,在单因素试验基础上,通过响应面优化盐析法脱除玛卡多糖中蛋白质的工艺条件。结果表明,盐析法脱蛋白最佳工艺为硫酸铵浓度4%、转速290r/min、盐析时间19min。在此工艺条件下脱除玛卡多糖中的蛋白质,蛋白质的清除率为(35.63±0.94)%,与理论值35.82%相比,误差0.19%;多糖回收率为(88.25±0.86)%,与理论值88.69%相比,误差0.44%。回归方程与实际情况拟合较好,表明优化工艺可靠。To determine the optimum process for removing protein from Maca polysaccharide by salting-out method.Based on the single factor experiment,the process conditions of removing protein from Maca polysaccharide by salting-out method were optimized by response surface methodology with the recovery rate of polysaccharide and the clearance rate of protein as the indexes.The results showed that the optimum process of deproteinization by saltingout method was ammonium sulfate concentration 4%,rotation speed 290r/min and salting-out time 19min.The removal rate of protein from Maca polysaccharide by this process was(35.63±0.94)%with an error of 0.19%compared with the theoretical value of 35.82%,and the recovery rate of polysaccharide was(88.25±0.86)%with an error of 0.44%compared with the theoretical value of 88.69%.The regression equation fits well with the actual situation,which shows that the optimized process is reliable.
