摘  要:建立高效液相色谱法测定食品中姜黄素类化合物含量的分析方法。利用单因素试验,对食品中姜黄素类化合物的提取试剂、提取方式、提取时间进行选择,确定最佳提取条件为甲醇振荡提取5 min。色谱条件:YMCTM C30色谱柱(150 mm×4.6 mm,5μm),流动相为乙腈-水(80∶20,v/v),用磷酸调节p H到3.0,流速1 m L/min,检测波长420 nm,柱温30℃。结果表明:3种姜黄素类化合物在0.10~10.00μg/m L内与峰面积有良好线性关系,生姜和咖喱中3种姜黄素类化合物的平均回收率分别为87.86%~107.35%和87.08%~107.47%,RSD均小于5%,3种姜黄素类化合物含量的相对标准偏差(RSD)均小于5%。该方法简单可行、重复性好、准确度高,可广泛用于食品中姜黄素类化合物含量测定。An analytical method was developed for determining total content of curcuminoids in food by high performance liquid chromatography. The extraction reagent,extraction method and extraction time of curcuminoids in food were selected by single factor experiment. Methanol extraction by shaking for 5 min was selected. The separation of curcuminoids were achieved on YMCTMC30 column( 150 mm ×4. 6 mm,5 μm) using acetonitrile-water( 80∶20,v/v) as the mobile phase at a flow rate of 1. 0 m L/min. The DAD detection wavelength and column temperature were set at 420 nm and 30 ℃,respectively. The results showed that the linear range of three curcuminoids were0. 10 ~ 10. 00 μg/m L. The average recovery rate of ginger and curry was 87. 86% ~ 107. 35% and 87. 08% ~ 107. 47% and RSD was less than 5%. The relative standard deviation( RSD) was no more than 5%. Therefore,this developed method is characteristics of simple operation,and high reproducibility and accuracy. It can be widely applied to determine total content of curcuminoids in food.
