摘  要:目的探究番茄红素能否抑制3T3-L1前脂肪细胞分化过程中脂质积累及其具体分子机制。方法利用MDI诱导前脂肪细胞分化为成熟脂肪细胞,并在前脂肪细胞分化过程中,给予0、12.5、25及50μM番茄红素共孵育8d,之后利用油红O染色分析不同浓度番茄红素对前脂肪细胞分化过程中脂质积累的抑制作用;Westernblots及RT-qPCR检测脂质代谢基因、p-AMPK、p-ACC以及线粒体呼吸链复合物I-IV表达,分析番茄红素对脂质代谢基因、线粒体脂肪酸β氧化以及线粒体功能的影响;用线粒体功能抑制剂寡霉素预先处理细胞4 h,之后番茄红素孵育8 d,Western blots检测FAS、p-ACC、PPARα及PPARγ表达,分析线粒体在番茄红素抑制前脂肪细胞分化过程中脂质积累所起作用。结果与MDI组相比,番茄红素可呈浓度依赖性抑制前脂肪细胞分化过程中脂质积累;可增加促脂质分解基因PPARα、PGC1ɑ及UCP1mRNA表达并减少促脂质积累基因FAS、PPARγ及C/EBPαmRNA表达;番茄红素促进了线粒体呼吸链复合物I-IV蛋白表达;促进了AMPK及ACC磷酸化;用寡霉素预先处理细胞,发现番茄红素对前脂肪细胞分化过程中脂质代谢基因表达的调节作用减弱。结论番茄红素可通过增加线粒体呼吸链复合物表达、促进AMPK及ACC磷酸化增强线粒体功能及脂肪酸β氧化分解,进而抑制前脂肪细胞分化过程中脂质积累。Objective To investigate the anti-adipogenic effects of lycopene(LYC)on lipid accumulation of differentiated 3 T3-L1 preadipocytes and the underlying molecular mechanisms.Methods 3 T3-L1 preadipocytes were incubated in an adipogenic differentiation MDI medium in the presence or absence of LYC(0,12.5,25 and 50μmol/L)for 8 day.Then,cells were stained with Oil Red O to elucidate the anti-adipogenic effects of LYC.Expressions of lipid metabolism genes,p-AMPK,p-ACC and mitochondrial respiratory chain complex I-IV were examined to detect the effects of LYC on lipid metabolism,mitochondrial fatty acidβ-oxidation and mitochondrial function.To further investigate the underlying connection between mitochondrial function and lipid metabolism,oligomycin,a mitochondrial function inhibitor(suppressing ATP synthesis)was employed in the study.Results LYC alleviated intracellular lipid accumulation in the process of 3 T3-L1 adipocyte differentiation in a dose-dependent manner.LYC increased the mRNA expressions of PPARɑ,PGC1ɑand UCP1 which were related to lipidolysis and decreased the mRNA expressions of FAS,PPARγand C/EBPɑwhich were related to lipid accumulation.Moreover,LYC enhanced mitochondrial function and fatty acidβ-oxidation through elevating the protein expressions of mitochondrial respiratory chain complex and promoting the phosphorylation of AMPK and ACC.Moreover,in the presence of oligomycin,the beneficial effects of LYC on lipid metabolism were partly removed in MDI-treated cells.Conclusion LYC suppressed MDI-triggered lipid accumulation through enhancing mitochondria function in adipocytes.
