摘  要:目的:探索Notch1与BPHL基因融合产物在胃癌组织中的表达和临床意义。方法:选取2018年5月至10月我院肿瘤外科50例初治、手术治疗且术后病理证实为胃癌患者的术后石蜡标本为研究对象,分别使用RT-PCR法及ARMS基因检测法检测胃癌组织、癌旁组织、转移淋巴结及正常胃黏膜中Notch1与BPHL基因融合产物的表达情况,分析其与不同临床病理类型的关系。结果:6例胃癌组织中检测到Notch1与BPHL基因融合产物,阳性率12.0%,癌旁组织和正常胃黏膜未检测到融合基因产物;性别间Notch1与BPHL基因融合产物检出阳性率比较差异无统计学意义(P>0.05);是否淋巴结转移组间Notch1与BPHL基因融合产物检测阳性率比较差异有统计学意义(P<0.05);不同分化程度组间Notch1与BPHL基因融合产物检测阳性率比较差异有统计学意义(P<0.05);不同浸润深度组间Notch1与BPHL基因融合产物检测阳性率比较差异有统计学意义(P<0.05)。结论:胃癌组织中Notch1与BPHL基因融合产物高表达可能参与胃癌的浸润及转移,对胃癌的诊断和治疗具有一定应用价值。Objective:To explore the expression and clinical significance of the fusion product of Notch1 and BPHL genes in gastric cancer tissues.Methods:The postoperative paraffin specimens of 50 patients with newly treated gastric cancer who underwent surgical treatment and confirmed by postoperative pathology were selected from our hospital.RT-PCR and ARMS gene detection methods were used to detect the expression of the fusion product of Notch1 and BPHL genes in gastric cancer tissues,adjacent tissues,metastatic lymph nodes and normal gastric mucosa and to analyze its relationship with different clinicopathological types.Results:The fusion product of Notch1 and BPHL genes was detected in 6 gastric cancer tissues,the positive rate being12%.The fusion gene product was not detected in adjacent tissues and normal gastric mucosa.There was no difference in positive rates of the fusion product of Notch1 and BPHL genes between different sexes(P>0.05).There was significant difference in positive rates of the fusion product of Notch1 and BPHL genes between metastatic lymph nodes and nonmetastatic lymph nodes(P<0.05).There was significant difference in positive rates of the fusion product of Notch1 and BPHL genes between differentiated tissues of different degrees(P<0.05).There was significant difference in positive rates of the fusion product of Notch1 and BPHL genes between infiltration tissues of different depths.Conclusion:The high expression of Notch1 and BPHLgene fusion product in gastric cancer tissue may be involved in the invasion and metastasis of gastric cancer,which is valuable in diagnose and treat gastric cancer to a certain degree.
