摘 要:以辣木叶水提取物为研究对象,利用氯仿、乙酸乙酯、正丁醇将辣木叶水提物进行分级,将不同质量浓度分级物作用于大鼠嗜碱性白血病细胞RBL-2H3,测定其类胰蛋白酶释放率、β-氨基己糖苷酶释放率、组胺释放率、细胞活力,并观察细胞形态变化。结果表明:辣木叶水提分级物均能提高RBL-2H3细胞类胰蛋白酶释放率,且与质量浓度呈正相关,其中氯仿分级物使RBL-2H3细胞类胰蛋白酶释放率最高增至27.11%;β-氨基己糖苷酶释放率与各分级物质量浓度呈正相关,其中氯仿分级物使RBL-2H3细胞β-氨基己糖苷酶释放率最高增至23.88%;氯仿分级物使RBL-2H3细胞组胺释放率最高增至94.00%;随分级物质量浓度的增加RBL-2H3细胞活力下降,随作用时间的延长RBL-2H3细胞活力下降。从细胞形态上看,氯仿分级物更易使RBL-2H3细胞脱颗粒,约12 h内能观察到脱颗粒。由此可见,辣木叶水提物中含有一定过敏成分,且可能集中于氯仿分级物中。该结果为探究辣木与健康,为辣木食品脱敏深加工提供一定数据支持。Moringa oleifera leaves water extracts was used as the research object.The water extract of Moringa oleifera leaves was graded with chloroform,ethyl acetate and n-butanol.The different mass concentration graded substance were applied to rat basophilic leukemia cells RBL-2H3,measure tryptase release rate,β-hexosaminidase release rate,histamine release rate,cell viability,and the changes of cell morphology were observed.The results showed that the water extracte graded substance of Moringa oleifera leaves can increase the tryptase release rate of RBL-2H3 cells,and it is positively correlated with the mass concentration.The chloroform graded substance can increase the tryptase release rate of RBL-2H3 cells to 27.11%.Theβ-hexosaminidase release rate is positively correlated with the mass concentration of each graded substance.The chloroform graded substance can increase theβ-hexosaminidase release rate of RBL-2H3 cells to 23.88%.The chloroform graded subtance increased the histamine release rate of RBL-2H3 cells to 94.00%.RBL-2H3 cell viability decreased with the increase of the mass concentration of the grading material,and the cell viability of RBL-2H3 decreased with the extension of the action time.In terms of cell morphology,the chloroform graded substance is more likely to degranulate RBL-2H3 cells,and degranulation can be observed within about 12 hours.It can be seen that the water extract of Moringa oleifera leaves contains certain allergens,which may be concentrated in the chloroform graded substance.The research provides certain data support for exploring Moringa oleifera and health,and for deep processing of Moringa food desensitization.
