摘 要:[目的]本研究旨在通过响应面分析法对醋酸杆菌发酵液多酚的提取工艺进行优化,并探究其对糙皮侧耳黄斑病的防治效果。[方法]以醋酸杆菌发酵液为供试材料,确定其多酚提取工艺中料液比、乙醇浓度、超声功率和超声时间的基础上,采用响应面分析法,对醋酸杆菌发酵液多酚的提取工艺条件进行优化。采用牛津杯法,确定醋酸杆菌发酵液多酚对黄斑病致病菌解淀粉芽孢杆菌的最小抑菌浓度,并进一步采用子实体涂抹抑制法,对醋酸杆菌发酵液多酚在糙皮侧耳子实体黄斑病的防治作用验证分析。[结果]醋酸杆菌发酵液多酚的最佳提取工艺条件为料液比1∶3、乙醇浓度45%、超声功率250 W和超声时间23 min,在此条件下多酚提取量为0.140 mg·mL^(-1)。牛津杯试验结果表明醋酸杆菌发酵液多酚对解淀粉芽孢杆菌有明显的抑制作用,且抑菌圈随着多酚浓度的降低而减小,最小抑菌浓度为17.5μg·mL^(-1)。子实体涂抹抑制试验结果进一步显示:醋酸杆菌发酵液多酚对黄斑病有明显抑制作用,但对糙皮侧耳子实体的正常生长无明显影响。[结论]本研究优化了醋酸杆菌发酵液多酚的提取工艺条件,同时结果表明醋酸杆菌发酵液多酚对糙皮侧耳黄斑病有明显防治作用,可为糙皮侧耳乃至其他大型真菌生产中细菌性病害的防治提供一个新的思路。[Objective]The present study was conducted to optimize the extraction process of Acetobacter zymotic fluid from phenolic(AZF)and to investigate its control effect on Bacillus amyloliquefaciens,the pathogenic bacteria of Yellow Blotch Disease(YBD)of Pleurotus ostreatus.[Methods]Based on the determination of solid-liquid ratio,ethanol concentration,ultrasonic power and ultrasonic time,response surface methodology was used to optimize the extraction conditions of AZF from phenolic.Then,the Oxford cup method was used to determine the minimum inhibitory concentration of phenolic from AZF against B.amyloliquefaciens.Further,the fruitbody smear inhibition method was used to further verify and analyze the prevention and control effect of phenolic from AZF on the fruitbody macular disease of P.ostreatus.[Results]The results showed that the optimal extraction process for AZF from phenolic was solid-liquid ratio at 1∶3,ethanol concentration at 45%,ultrasonic power at 250 W,and ultrasonic time at 23 min.Later,Oxford cup test results showed that phenolic from AZF had significant inhibitory effect on B.amyloliquefaciens,and the inhibition zone decreased with the decrease of phenolic concentration,and the minimum inhibitory concentration was 17.5 ug·mL^(-1).The results of the fruiting body smear inhibition test showed that the phenolic from AZF had obvious inhibitory effect on YBD,but had no obvious effect on the normal growth of the fruiting body of P.ostreatus.[Conclusion]In this study,the extraction conditions of AZF from phenolic were optimized,and the results showed that the phenolic had obvious inhibitory effect on B.amyloliquefaciens,which provided an important reference for the biological control of P.ostreatus.
