摘 要:通过改良阿斯贝(Ashby)无氮培养基和乙炔还原法从牛蒡根内分离筛选出固氮能力较强的菌株,通过形态观察及分子生物学技术对其进行鉴定,并通过响应面法对其培养条件进行优化。结果表明,筛选出一株固氮能力较强的菌株NF-112,初步鉴定为肺炎克雷伯氏菌(Klebsiella pneumoniae),其最优培养条件为初始pH值5.3、培养温度28℃、葡萄糖添加量26 g/L。在此最优培养条件下,OD600 nm值为1.76。该研究为药用植物牛蒡专用生物有机肥制备奠定理论和技术基础。The strains with strong nitrogen fixation ability were isolated from the burdock root by improved Ashby nitrogen free medium and acetylene reduction method,and identified by morphological observation and molecular biology techniques.The culture conditions of the strain were optimized by response surface methodology.The results showed that a strain numbered NF-112 with strong nitrogen fixation ability was screened and preliminarily identified as Klebsiella pneumoniae.The optimal culture conditions of the strain were obtained as follows:initial pH 5.3,culture temperature 28℃,glucose addition 26 g/L.Under the optimal culture conditions,the OD600 nm value was 1.76.The study laid a theoretical and technical foundation for the special bio-organic fertilizer preparation of the medicinal plant burdock.
