摘  要:以瓦尼木层孔菌为原料,菌丝生物量为评价指标,响应面法优化发酵培养基的碳源和氮源,通过单因素试验,确定最佳发酵工艺,为瓦尼木层孔菌的开发利用提供理论参考。结果表明,发酵培养基的最佳碳源和氮源及其添加量是蔗糖2%、可溶性淀粉1.5%、酵母浸粉0.5%、蛋白胨0.5%,最佳发酵条件是液体种子培养时间3d、培养温度26℃、初始pH7、摇瓶转速160r/min、接种量15%、摇瓶装液量80mL/250mL,在此条件下进行验证试验,菌丝生物量为1.1780g/100mL。Using Phellinus vaninii as the raw material,mycelium biomass was used as an evaluation index,carbon and nitrogen sources for fermentation medium were optimized by response surface methodology.The best fermentation process was determined by single factor experiment.The study provided a theoretical reference for comprehensive utilization of Phellinus vaninii.The results showed that the best carbon and nitrogen sources of fermentation medium and its addition amount were sucrose 2%,soluble starch 1.5%,yeast 0.5%,and peptone 0.5%.The best fermentation conditions were liquid seed culturing for 3 days,at 26℃,initial pH 7,rotation shaker’s speed 160r/min,inoculum amount 15%,and flask loading volume 80 mL/250 mL.The validation test under this conditions showed that the mycelium biomass was 1.1780g/100mL.
