摘　　要：目的研究SiRNA沉默β-catenin对MGC803细胞Wnt-5a基因表达的影响。方法构建RNA干扰质粒,经脂质体转染胃癌MGC-803细胞。将细胞分为质粒组(转染)和空白组(未转染)。检测β-catenin表达情况用RT-PCR技术;检测Wnt-5a含量用免疫组化SP染色法。结果β-catenin质粒组OD值(0.7398±0.036),空白组OD值(1.6835±0.023),差异有统计学意义(P<0.01)。Wnt-5a免疫组化染色观察到质粒组细胞质染色呈浅黄色,质粒组与空白组相比,细胞质着色明显减弱,黄染区域变小。结论通过RNAi干扰,MGC-803细胞β-catenin表达下调,质粒组的Wnt-5a表达减弱。Objective To studyβ-catenin SiRNA to the gastric carcinoma MGC-803 Wnt-5a gene ex-pression of the influence.Methods Build SiRNA interference plasmid,through liposome transfected into MGC-803 cells.Divides the cell into two groups:the plasmid interference group(transfected)and the blank control group(not transfected).RT-PCR technology detect the expression ofβ-catenin;the two control groups were detected in the Wnt-5a content by Immunohistochemical SP staining method.Resultsβ-catenin OD value of plasmid in-terference groups(0.7398±0.036),β-catenin OD value of blank control group(1.6835±0.023),the compari-son between two groups difference have statistically significant(P<0.01).Immunohistochemical staining Wnt-5a showed that,plasmid interference group compared with the blank control group cytoplasm shading weakened dra-matically,yellow dye regional decrescent.Conclusion Through RNAi technology silence,β-catenin expression is down-regulated in MGC-803;Wnt-5a gene of the interference plasmid group expression is the weaker.
