摘  要:目的探讨多发性特发牙颈部外吸收患者TNFRSF11A基因突变情况。方法本研究纳入2例MICRR患者、1例家系成员及5例健康者为研究对象。外周血中提取所有研究对象基因组DNA,应用聚合酶链式反应对其TNFRSF11A基因1号外显子进行测序。测序结果与GenBank公布的正常序列比对,寻找有无突变。结果多发性特发牙颈部外吸收患者、家系成员及健康者的目标基因位点上均无突变发生。结论多发性特发牙颈部外吸收患者TNFRSF11A基因1号外显子无突变,排除其为该疾病致病基因的可能性。Objective To investigate the mutation of TNFRSF11 A gene in patients with multiple idiopathic cervical root resorption(MICRR).Methods A total of two cases of MICRR,one family member and five normal persons were examined in this study.Genomic DNA was extracted from the blood samples.Polymerase chain reaction(PCR)was performed to amplify exon 1 of the TNFRSF11 A gene and the PCR products were directly sequenced to detect the mutation.The sequence results were compared to the normal sequence of GenBank.Results There were no mutations in the target gene loci in patients,family member and normal persons.Conclusion There was no mutation in exon 1 of TNFRSF11 A gene in patients with MICRR,thus ruling out the possibility of TNFRSF11 A as the pathogenic gene of this disease.
