摘　　要：目的观察结肠癌患者肿瘤组织中miR-383、Notch1 mRNA的表达情况,并分析其与临床病理参数及预后的关系。方法收集结肠癌患者126例,采用qRT-PCR法检测结肠癌组织和癌旁正常组织中miR-383、Notch1 mRNA的表达情况,分析结肠癌组织中miR-383与Notch1 mRNA表达的相关性及其与患者临床病理参数的关系。随访5年,以miR-383、Notch1 mRNA表达量中位数为界限,将患者分为miR-383、Notch1 mRNA高表达者与低表达者,Log-Rank检验比较其生存时间。结果结肠癌组织中miR-383相对表达量低于癌旁组织,Notch1 mRNA相对表达量高于癌旁组织(P均<0.01);结肠癌组织miR-383、Notch1 mRNA相对表达量呈负相关关系(r=-0.665,P<0.01)。与Ⅲ~Ⅳ期、有淋巴结转移患者比较,TNM分期Ⅰ~Ⅱ期、无淋巴结转移患者结肠癌组织miR-383相对表达量增高、Notch1 mRNA相对表达量降低(P均<0.05);不同性别、年龄、肿瘤直径、分化程度患者的miR-383、Notch1 mRNA相对表达量比较差异均无统计学意义。miR-383高表达者5年生存率高于低表达者,Notch1 mRNA高表达者5年生存率低于低表达者(P均<0.05)。结论结肠癌组织中miR-383低表达,Notch1 mRNA高表达,且两者表达量与TNM分期、淋巴结转移及预后相关,有望成为新的结肠癌诊断和预后判断的分子标志物。Objective To investigate the expression levels of microRNA(miRNA)-383 and Notch1 mRNA in the colon cancer tissues and their relationships with clinicopathological parameters and prognosis.Methods The clinical data of 126 patients with colon cancer were collected,and the expression levels of miR-383 and Notch1 mRNA in the cancer tissues and adjacent tissues were detected by qRT-PCR.Pearson correlation was used to analyze the correlation between miR-383 and Notch1 mRNA expression in the colon cancer tissues,and we analyzed the relationships between the two and the clinicopathological characteristics of patients.After follow-up for 5 years,the patients were divided into patients with high miR-383 and Notch1 mRNA expression and those with low expression by taking the median of miR-383 and Notch1 mRNA expression as the limit,and their survival time was compared by Log-Rank test.Results The expression of miR-383 in the colon cancer tissues was significantly lower than that in the adjacent tissues,and the expression of Notch1 mRNA was significantly higher than that in adjacent tissues(both P<0.01).The miR-383 expression was negatively correlated with Notch1 mRNA expression in the colon cancer tissues(r=-0.665,P<0.01).The relative expression of miR-383 in the colon cancer tissues of patients with TNM stagesⅠ-Ⅱand no lymph node metastasis was higher than that of patients with stageⅢ-Ⅳand patients with lymph node metastasis;the relative expression of Notch1 mRNA was lower than that of patients with stageⅢ-Ⅳand patients with lymph node metastasis(all P<0.05).There was no statistically significant difference in the relative expression of miR-383 or Notch1 mRNA between patients with different gender,age,tumor diameter,and degree of differentiation.The 5-year survival rate of patients with high expression of miR-383 was higher than that of patients with low expression,and the 5-year survival rate of patients with high expression of Notch1 mRNA was lower than that of patients with low expression(both P<0.05).Conclus
