摘 要:目的:利用Plackett-Burman和Box-Behnken效应面法优化椒目仁油微波-超声辅助提取工艺并研究其降血脂作用。方法:建立α-亚麻酸的气相测定方法,以椒目仁油的提取率和α-亚麻酸含量总评值为指标,筛选椒目仁油最佳提取工艺;建立大鼠高血脂模型评价其降血脂能力。结果:最佳工艺为椒目药材粉碎粒径65目,8倍正己烷,微波功率392 W处理83 s,超声功率297 W处理9 min,搅拌速度90 r/m,在此条件下椒目仁油提取率为17.41%,α-亚麻酸的含量为32.05%,所得椒目仁油能降低模型大鼠血清中总胆固醇(TC)、甘油三酯(TG)和低密度脂蛋白(LDL-C)水平,升高其血清高密度脂蛋白(HDL-C)水平。结论:本实验优选的椒目仁油提取工艺方法可行,制备的椒目仁油具有较好的降血脂作用。Objective:Design of Plackett-Burman(PB)and Box-Behnken(BB)were used to optimize the Microwave-Ultrasound extraction technology of zanthoxylum seed oil(ZSO)and observe the effect of hyperlipidemia in rats.Methods:The gas chromatography(GC)method was established to determine the content ofα-linolenic acid.By analyzing PB experimental result,ultrasonic time,ultrasonic temperature and ultrasonic power were optimized by response surface methodology.To establish a rat model of hyperlipidemia and evaluate its ability of reducing blood lipid.Results:Optimal extraction conditions were as follows:particle size of 65 meshes,8 times n-hexane,microwave time 83 s and ultrasonic power of 392 W,ultrasonic time 9 min and ultrasonic power of 297 W,and stirring speed 90 r/m,the extraction rate of ZSO was 17.41%and the content ofα-linolenic acid was 32.05%.The obtained ZSO could reduce the serum level of TC,TG,LDL-C and increase the HDL-C level.Conclusion:The extraction technology of ZSO optimized by experiment is feasible,and the ZSO has a better effect on reducing blood lipid.
