摘 要:为比较银耳多糖、透明质酸对D-半乳糖致衰小鼠皮肤的改善作用及相应机理,将SPF级KM雄性小鼠随机分为Normal(正常组)、Model(模型组)、透明质酸组(HA,100mg/kg·BW)和银耳多糖组(TFP,100mg/kg·BW),除Normal外其余3组每天腹腔注射D-半乳糖120mg/kg同时灌喂样品连续8周。实验结束后处死小鼠,取背部皮肤测羟脯氨酸、透明质酸含量,检测各组小鼠组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活力及丙二醛(MDA)浓度以及肠道内pH和短链脂肪酸(short chain fatty acids,SCFAs)含量。结果发现透明质酸和银耳多糖可提升衰老皮肤中羟脯氨酸和透明质酸含量(p <0.01),提高血清、肝脏和心脏部位GSH-Px、SOD活性并降低MDA浓度(p <0.01),两者效果无统计学差异。此外,透明质酸、银耳多糖逆转了D-半乳糖所致小鼠结肠内pH下降和SCFAs浓度增加,特别是正丁酸和异丁酸,表明对肠道菌群具有调节作用。由此提示,透明质酸和银耳多糖对小鼠衰老皮肤的改善作用可能是通过提高机体抗氧化酶活性实现。To compare the improvement effects of hyaluronic acid(HA)and Tremella fuciformis polysaccharides(TFP)on aging skin texture in D-galactose induced ageing mice and to explore the underlying mechanism.Methods :SPF-grade KM male mice were randomly divided into four groups :Normal(given physiological saline),Model(given physiological saline),HA(oral administration of HA 100 mg/kg · BW),and TFP(oral administration of TFP 100 mg/kg · BW). The Normal group were intraperitoneally injected with physiological saline,and the remaining groups were intraperitoneally injected with a dosage of 120 mg/kg · BW D-galactose per day for eight-week. All mice were killed at the end of the experiment. Then,the contents of hydroxyproline and HA in the skin,the activities of GSH-Px,SOD and the concentrations of MDA in serum,liver and heart tissues,and the pH and the concentrations of SCFAs in intestinal tracts were measured. Both HA and TFP increased the contents of hydroxyproline and hyaluronic acid in ageing skin(p < 0.01)equivalently. Further,HA and TFP improved the activities of GSH-Px and SOD and reduced the concentrations of MDA(p < 0.01)in serum,liver and heart tissues substantially. Additionally,there was no statistical difference between the effects of the two. Compared with normal group,the pH decreased while the concentrations of total SCFAs increased in the colon in the Model group. Interestingly,HA increased the pH but reduced the concentrations of SCFAs obviously in the colon in aging mice,especially the concentrations of N-butyric acid and isobutyric acid. Similarly,TFP reversed the increases in pH and the alterations in the concentration of SCFAs in the colon in ageing mice caused by D-galactose treatment.Collectively,HA and TFP may improve the ageing skin texture through increasing antioxidant enzyme activities in ageing mice.
