摘  要:旨在探究超声辅助复合酶法提取黄精多糖的最佳工艺,并研究其体外抗氧化活性,以黄精多糖得率为响应值,以液料比、加酶量、超声功率和酶解pH值4个因素为自变量,通过Box-Behnken中心组合方法设计试验进行响应面分析,确定最佳工艺参数;通过DPPH自由基清除能力、超氧自由基清除能力测定黄精多糖的抗氧化活性。结果表明:黄精多糖最佳工艺参数液料比(mL/g)为21∶1,加酶量1.6%,超声功率360 W,酶解pH值5.0,此条件下黄精多糖得率最高,为27.02%,与响应面预测值27.81%相近;抗氧化试验表明,黄精多糖对DPPH自由基和超氧自由基的半清除率(IC;)分别为1.08 mg/mL、0.93 mg/mL,且其对自由基清除能力随着其浓度的增加而增大。研究结果为黄精多糖的开发和利用提供了参考依据。In order to explore the optimal extraction process for polysaccharide from Ploygonatum sibiricum by ultrasound-assisted complex enzyme method and to determine its antioxidant activity in vitro, the yield of Ploygonatum sibiricum polysaccharide was used as the response value, and the liquid-solid ratio, enzyme dosage, ultrasonic power and enzymatic hydrolysis pH were used as the independent variables. Through the box Behnken central composite design test, response surface analysis was carried out to determine the optimal process parameters. The antioxidant activity of Ploygonatum sibiricum polysaccharide was determined by DPPH free radical scavenging ability and superoxide free radical scavenging ability. The results showed that the optimum technological parameters of Ploygonatum sibiricum polysaccharide were as follows: the ratio of liquid to material was 21∶1(mL/g), the amount of enzyme was 1.6%, the ultrasonic power was 360 W, and the enzymatic hydrolysis pH was 5.0. Under these conditions, the yield of Ploygonatum sibiricum polysaccharide was the highest, which was 27.02% which was close to 27.81% predicted with the response surface method. The results of the antioxidant test showed that the half scavenging rates(IC;) of Ploygonatum sibiricum polysaccharid on DPPH free radical and superoxide free radical were 1.08 mg/mL and 0.93 mg/mL, respectively. The free radicals scavenging ability of Ploygonatum sibiricum polysaccharide increased with the increase of its concentration.The present results provided reference for development and utilization of Ploygonatum sibiricum polysaccharide.
