摘 要:通过建立高效、准确、适宜批量测定甘薯块根多酚含量的方法,为筛选评价高多酚含量甘薯种质提供技术支持。对提取溶剂、福林酚、碳酸钠等浓度及反应时间进行系统优化,并对建立的甘薯多酚高效检测方法的稳定性、可靠性进行评价;采用优化的方法对国内83个甘薯品种的多酚含量进行测定。结果表明,以乙醇做提取剂,最佳体积分数为70%,最佳福林酚体积分数为15%,碳酸钠质量分数为5%,最佳反应时间60 min;采用酶标仪及96孔板可同时检测30份样品;该方法的平均加标回收率为98.4%,3次重复测定相对标准偏差为2.4%,表明建立的方法准确度高,重复性好;83份甘薯样品的多酚质量分数在0.22~2.00 mg/g鲜样之间,且紫肉品种中的多酚含量显著高于其他品种。The purpose of this study was to establish an efficient,accurate,and suitable method for batch determination of polyphenol content in sweet potato tubers,and to provide technical support for screening and evaluating sweet potato varieties with high polyphenol content.The concentration of extraction solvent,Folin-ciocalteu,Na2CO3,and the reaction time were systematically optimized,and the stability and reliability of the established high-efficiency detection method for sweet potato polyphenol were evaluated.The optimized method was used to determine the polyphenol content of 83 domestic sweet potato varieties.The results showed that the optimal concentration of ethanol as extractant was 70%,and the optimum concentration of the Folin-ciocalteu was 15%.The concentration of Na2CO3 was 5%.The reaction time was 60 min.Using microplate reader and 96-well plate could detect 30 samples simultaneously.The average spike recovery of this method was 98.4%,and the relative standard deviation of three repeated determinations was 2.4%,indicating that the established method had high accuracy and good repeatability.The polyphenol content of 83 sweet potato samples was 0.22~2.00 mg/g fresh samples,and the polyphenol content in purple sweet potatoes varieties was significantly higher than that in other varieties.
