摘  要:以航天诱变胶红酵母突变株JH-R23为研究对象,通过单因素和响应面法对胶红酵母突变株JH-R23合成虾青素培养基进行优化,同时对其生长特性进行探究。结果得到胶红酵母突变株JH-R23合成虾青素最优培养基配方为:葡萄糖41.24 g/L、蛋白胨5.79 g/L、硫胺素5.17 mg/L、磷酸氢二钾1 g/L、七水合硫酸镁0.5 g/L、氯化钾0.5 g/L、硫酸亚铁0.01 g/L,pH值为6.27;合成虾青素较适宜温度在23~33 ℃。通过培养基优化,胶红酵母突变株JH-R23虾青素产量比优化前提高2.18倍,且虾青素合成为典型生长偶联型,生产温度范围广、产量高,使该菌株有望成为工业化生产虾青素的优良菌种。The space mutation of Rhodotorula mucilaginosa JH-R23 was studied.The astaxanthin synthesis medium of Rhodotorula mucilaginosa mutant JH-R23 was optimized by single factor and response surface methodology,and its growth characteristics were explored.The optimal medium for astaxanthin synthesis of Rhodotorula mucilaginosa mutant JH-R23 was glucose 41.24 g/L,peptone 5.79 g/L,thiamine 5.17 mg/L,dipotassium phosphate 1 g/L,magnesium sulfate heptahydrate 0.5 g/L,potassium chloride 0.5 g/L,ferrous sulfate 0.01 g/L,pH 6.27;The optimum temperature for astaxanthin synthesis is 23~33 ℃.Through the optimization of medium,the astaxanthin yield of Rhodotorula mucilaginosa mutant JH-R23 was 2.18 times higher than that before optimization,and the astaxanthin synthesis is a typical growth coupling type,has wide production temperature range and high yield,these advantages make it a good strain for industrial production of astaxanthin.
